摘 要:目的:研究地尔硫芯对葛根素血药浓度及药动学参数的影响。方法:应用高效液相色谱法测定与地尔硫芯合用前后葛根素的血药浓度。结果:注射合用后,地尔硫芯在低剂量时(0.5 mg·kg-1)对葛根素的药动学过程无显著性影响(P>0.05),但在高剂量时(2.5 mg·kg-1)有显著性影响(P<0.05)。结论:葛根素与地尔硫芯合用时,在一定浓度范围内,地尔硫芯可抑制葛根素从大鼠体内的消除;二者合并用药时应对葛根素进行临床给药监测。OBJECTIVE To study the effect of diltiazem on serum concentration and pharmacokinetics of puerarin in rats. METHODS The concentration of puerarin in serum was detected by high performance liquid chromatography (HPLC) before and after diltiazem administration. RESULTS There was no significant difference was found between the control and 0. 5 mg. kg-1 diltiazem combined groups for intravenous administration (P〉0.05), and there was significant difference between the control and 2. 5 mg. kg-1 diltiazem combined groups (P〈0. 05). CONCLUSION Diltiazem inhibited puerarin metabolism when puerarin was coadministered with diltiazem, so it is necessary to change the therapeutic dose of puerarin.
