摘 要:目的:建立一种快速﹑准确和标准化的龙胆科"地格达"类蒙药材的DNA条形码鉴别方法,为"地格达"类蒙药材的分子鉴定提供依据。方法:对蒙药地格达类药用植物进行通用引物PCR扩增,PCR扩增产物直接测序,并对序列进行分析。结果:测得4种"地格达"的rbcL,ITS,trnH-psbA和matK全长序列,4种序列长度的范围为388~940 bp,rbcL,ITS,matK序列都可以将其鉴别出来。结论:trnH-psbA序列不适合用于4种"地格达"的鉴别,ITS序列可作为地格达类蒙药材一种良好的分子标记。通过DNA条形码鉴别方法可对4种龙胆科"地格达"进行准确﹑可靠﹑有效的分子鉴别。Objective: To establish a rapid, accurate and standardized method for identifying Gentianaceae mongolian medicine Digeda with DNA barcoding method, and to provide the basis of molecular authentication of mongolian medicine Digeda. Method : The mongolian medicine Digeda was amplified by PCR with universal primer and sequenced directly, the sequence was aligned and analyzed. Result: The complete rbcL, ITS, trnH-psbA and matK sequence of four Digeda species have been obtained, and the length of four sequences region varied from 388 to 940 bp. They were distinguished by rbcL, ITS or matK sequence. Conclusion: The trnH-psbA sequence was not suitable to identify the four species of Digeda. The ITS region could be a pretty good marker to distinguish mongolian medicine Digeda. DNA barcoding identification method is accurate and reliable and can be applied to identify the four kinds of Gentianaceae Digeda-species mongolian medicine materials.
