摘 要:目的建立蒸发光散射法测定血浆中福多司坦含量的方法。方法色谱柱:Brava BDS C8柱(250mm×4.6mm,5μm);流动相:甲醇-水(2∶98);流速:1.0mL.min-1;柱温:40℃;漂移管温度:110℃,载气(N2)流速:2.5L.min-1。结果在选定的色谱条件下,测定的线性范围为0.2~30μg.mL-1,r=0.999 6,定量下限为0.2μg.mL-1,平均相对回收率为96.3%,日内与日间的RSD值均小于10%。结论该方法准确、精密、灵敏度高、简便、快速,适用于血浆福多司坦质量浓度的测定。Objective To establish a simple HPLC-ELSD method for the determination of fudosteine in human plasma.Method The separation was performed on a Brava BDS C8 column(250 mm×4.6 mm,5 μm) at 40 ℃.The mobile phase was a mixture of methanol-water(2∶98) and the flow rate was 1.0 mL·min^-1,An evaporative light scattering detector(ELSD) was connected to HPLC with the drift tube temperature of 110 ℃ and nitrogen gas flow rate of 2.5 L·min^-1.Results Under the selected chromatographic conditions,the calibration curves had good linearity in the range of 0.2-30 μg·mL-1(r=0.999 6).The lower limit of quantification was 0.2 μg·mL-1,and the average recovery was 96.3%.The within-day and between-day RSD were less than 10%(n=5).Conclusion This method was accurate,sensitive,simple and rapid,and it could be applied in the determination of fudosteine in human plasma.
