摘 要:目的 :研究糖皮质激素诱导不同发育阶段培养的大鼠睾丸间质细胞凋亡状况。方法 :以Annexin V FITC和PI双标细胞 ,应用激光共聚焦显微镜和流式细胞仪检测。 结果 :ILC、ALC经皮质酮处理后 ,其凋亡量显著高于对照组 ;PLC经皮质酮处理后 ,其凋亡量与对照组无显著差异。 结论 :糖皮质激素能诱导大鼠睾丸未成熟型和成熟型间质细胞凋亡 ,而不能诱导前体型间质细胞凋亡。WT5”HX]Objectives:The possibility of glucocorticoid mediated induction of apoptosis was investigated in rat cultured Leydig cells of 3 distinct stages of differentiation. [WT5”HX]Methods: Apoptotic Leydig cells double labeling with Annexin and PI were observed by confocal laser scanning microscope( CLSM) and flow cytometry(FCM),respectively. [WT5”HX]Results: Both CLSM and FCM analysis of Annexin V FITC labeling progenitor Leydig cell (PLC) showed that PLC with glucocorticoid treatment didn't reveal a significant increase of apoptosis compared to control.CLSM analysis of Annexin V FITC labeling showed that both immature Leydig cell( ILC) and adult Leydig cell(ALC) exposed to glucocorticoid had a significantly increased frequencies of apoptosis.Increased ILC and ALC apoptosis was also found by FCM. [WT5”HX]Conclusions: These results suggested that glucocorticoid can initiate the process of apoptosis of rat cultured ILC and ALC,thereby resulting in decline in testosterone production.
