摘 要:目的观察盐酸戊乙奎醚(PHC)对脓毒血症(CLP)大鼠外周血内皮细胞(CECs)及其微粒(EMPs)计数的影响。方法 120只SD雄性大鼠随机分成六组:①对照组(sham组):假手术+生理盐水2 mL;②CLP组:CLP+生理盐水2 mL;③PHC0.1组:CLP+PHC 0.1 mg/kg;④PHC0.5组:CLP+PHC 0.5 mg/kg;⑤PHC1.0组:CLP+PHC 1.0 mg/kg;⑥PHC2.0组:CLP+PHC 2.0 mg/kg。方法流式细胞学(FCM)检测大鼠CECs及EMPs计数,ELISA法检测血浆TNF-α、IL-6,基质显色鲎法检测血浆内毒素(LPS)水平。每组随机各抽取10只分别观察大鼠6、12、24、48 h以及72 h生存率。结果同时点比较各组生存率无差异;TNF-α、IL-6水平和外周血CECs和EMPs计数:使用盐酸戊乙奎醚各组均低于CPL组,盐酸戊乙奎醚1.0 mg/kg和2.0 mg/kg组(两组间比较差异无统计学意义)低于0.1 mg/kg与0.5 mg/kg组(两组间比较差异无统计学意义);sham组LPS含量低于所有其他组,而其他各组间LPS含量比较差异无统计学意义。结论盐酸戊乙奎醚可明显减少CLP大鼠CECs及EMPs计数,减轻脓毒血症导致的血管内皮损伤,其可能机制是通过抑制炎症反应及炎症因子产生与释放实现。Objective To investigate effects of Penehyclidine Hydrochloride on the counts of circulating endothelial cells(CECs) and endothelial micropaticles(EMPs) in sepsis rats.Methods One hundred and twenty rats were randomly divided into 6 group: sham group(sham operation + Saline 2 mL),CLP group(CLP+ Saline 2 mL),PHC 0.1 group(CLP+PHC 0.1mg/kg),PHC0.5 group(CLP+PHC 0.5 mg/kg),PHC1.0 group(CLP+PHC 1 mg/kg),PHC2.0 group(CLP+ PHC 2 mg/kg.CECs and EMPs counts and the level of TNF-α,IL-6,LPS in sepsis rat were detected by flow cytometry or enzyme-linked immunosorbent assay.Survival rate in 6,12,24,48,72 h between the six groups were measured.Results No deference of survival rate among CLP group,PHC0.1 group,PHC0.5 group,PHC1.0 group and PHC2.0 group was found.The level of TNF-α,IL-6,LPS and the counts of circulating endothelial cells(CECs) and endothelial micropaticles(EMPs) in groups which PHC had been used in were lower than CPL group,all of above in PHC1.0 group and PHC2.0 group(no deference was found between the two groups) were lower than that in PHC0.1 group and PHC0.5 group(no deference was found between the two groups),no deference of LPS among CLP group,PHC0.1 group,PHC0.5 group,PHC1.0 group and PHC2.0 group was found.Conclusion Penehyclidine Hydrochloride can reduce the number of circulating endothelial cells(CECs) and endothelial micropaticles(EMPs) in sepsis rats,extenuate injury of endothelial cells and the protective effects may be related to antioxidation and inhibition of inflammatory response.
