摘　　要：目的 :探讨L 精氨酸对体外培养的人肾系膜细胞 (HMCL)增殖的影响。方法 :用四甲基偶氮唑盐比色法测定不同浓度L 精氨酸培养液中HMCL的增殖情况。结果 :随L 精氨酸浓度增加 ,细胞生长受抑制。L 精氨酸浓度大于 2 0mmol·L-1时 ,细胞生长的抑制与对照组比较有统计学意义。选择L 精氨酸敏感浓度 2 5mmol·L-1评价其时间效应 ,0～ 48h内抑制率增加。结论 :L 精氨酸抑制人肾系膜细胞增殖 ,呈浓度、时间依赖性 ,其对治疗慢性肾纤维化具有潜在价值。Objective The experiment was designed to study the effect of L ?arginine on the proliferation of human mesangial cells in vitro by using 3?(4,5?dimethylthiazole?2?yl)?2,5?diphenyl tetrazolium bromide(MTT) method. Methods The effects of L ?arginine on human mesangial were cells examined by cell culture technology and the method of MTT.Result Cell growth was significantly inhibited when the concentration of L ?arginine in the media was more than 20?mmol·L -1 compared with the control group, which showed dose?dependent manner. Furthermore, the antiproliferative effect of L ?arginine was demonstrated as time?dependent manner as well in this experiment,inhibitive rate increasing in 0～48?h at 25?mmol·L -1 . Conclusion L ?arginine might exert an antiproliferative effect on the human renal mesangial cells with a dose?and time?dependent manner.
