替米沙坦对糖尿病大鼠肾脏组织中TSP-1的影响

摘　　要：目的探讨替米沙坦对糖尿病大鼠肾组织中血小板反应蛋白-1(TSP-1)表达的影响。方法采用一次性腹腔注射链脲佐菌素60 mg/kg建造大鼠糖尿病模型。将实验动物随机分为正常对照组(A组)、糖尿病未干预组(B组)和糖尿病替米沙坦干预组(C组)3组。C组给予替米沙坦10/mg.kg-1.d-1灌胃,A组和B组给予等剂量生理盐水灌胃作对照。12周后,测定各组大鼠血尿素氮(BUN)、血肌酐(Scr)、24 h尿蛋白及肾脏肥大指数等肾脏功能指标;R T-PCR法和免疫组化法检测肾脏TSP-1 mR NA及其蛋白的表达水平。结果与A组相比,B组和C组24 h尿蛋白和肾脏肥大指数显著增高(P<0.05或0.01);与B组相比,C组24 h尿蛋白和肾脏肥大指数显著降低(P<0.05)。与A组相比,B组和C组TSP-1表达显著增高(P<0.01);与B组相比,C组TSP-1表达显著降低(P<0.01);TSP-1蛋白表达水平与24 h尿蛋白、肾脏肥大指数均呈显著正相关(P<0.01)。结论 AR B类药物对糖尿病大鼠具有肾脏保护作用,其机制可能部分与降低了肾组织内TSP-1的表达有关。【Objective】 To investigate the effect of telmisartan on the expression of Thrombospondin-1（TSP-1） in the kidneys of diabetic rats.【Methods】 A rat model of diabetes was induced by a single tail intravenous injection of streptozotocin 60 mg/kg.Experimental animals were randomly assigned to 3 groups： normal control（group A）,diabetes（group B） and DM treated with telmisartan（group C）.The latter group was administered with telmisartan（10 mg/kg-1·d-1）,the remainder received the same volume vehicle.12 weeks after treatment,renal function including Blood Urea Nitrogen（BUN）,Serum creatinine（Scr）,24 hour Urine protein quantity and renal mass index were tested.The expression of TSP-1 mRNA was detected by reverse transcriptio1n polymerase chain reaction（RT-PCR） and expression of TSP-1 protein was determined by immunohistochemistry.【Results】 The 24-hour Urine protein quantity and renal mass index in group B and group C were significantly higher than group A（P 〈0.05 or 0.01）.And the 24-hour Urine protein quantity and renal mass index in group C showed a significant decrease compared with group B（P 〈0.05）.The expression of TSP-1 was significantly increased in the kidneys of group B and group C,compared with group A（P 〈0.01）.TSP-1 expression in group C was lower than group B（P 〈0.01）.The expression level of TSP-1 had positive relationship with 24 h uria protein and renal mass index（P 〈0.01）.【Conclusion】 Our study indicates that telmisartan treatment could mildly protect kidneys of diabetic rats by inhibition of TSP-1 expression.
