摘 要:目的观察黄芩苷衍生物对大鼠心肌缺血再灌注致心肌损伤后心功能及心肌梗死的保护作用。方法将60只雄性Wistar大鼠随机平均分为6组,即假手术组、模型组、维拉帕米0.5mg/kg组、黄芩苷衍生物45、90、180 mg/kg组。采用结扎冠状动脉左前降支30 min,再灌注120 min的方法建立大鼠心肌缺血再灌注损伤模型,分别于缺血前及再灌注前10 min内通过股静脉注射黄芩苷衍生物(45、90、180 mg/kg)。于缺血前、缺血30 min及再灌注30、60、90、120 min时,分别测定左心室收缩压(LVSP)、左心室内压最大上升速率(+dp/dt max)、左心室最大下降速率(-dp/dt max)、心率、收缩压、舒张压及平均动脉压等指标,以评价心脏功能。并用TTC染色法测定大鼠心肌梗死范围。结果黄芩苷衍生物能够显著对抗心肌缺血再灌注损伤引起的LVSP、±dp/dt max及血压的下降,抑制心率过度减慢。剂量依赖性地缩小心肌梗死范围即心肌梗死比例(梗死区重/左心室重):模型组、黄芩苷衍生物45、90、180 mg/kg组的梗死范围分别为(33.1±6.0)、(29.9±5.4)、(24.8±5.0)及(22.3±3.4)(均为P<0.05)。结论黄芩苷衍生物对大鼠心肌缺血再灌注损伤有保护作用。Objective To evaluate the protective effects of Baicalin derivant on acute myocardial ischemia reperfusion injury in rats. Methods Sixty male Wistar rats were randomly divided into six groups:Sham group,model group,verapamil group(0.5 mg/kg) and different Baicalin derivant group(45,90 and 180 mg/kg).Myocardial ischemia reperfusion injury rat model was made by ligating left anterior descending coronary artery for 30-min followed by 120 min of reperfusion.Baicalin derivant was given intravenously before ischemia and 10 min before reperfusion.Left ventricular systolic pressure(LVSP),maximal rate of rise of ventricular pressure(+dp/dt max),maximal rate of decline of ventricular pressure(-dp/dt max),heart rate(HR),systolic blood pressure(SBP),diastolic blood pressure(DBP) and mean arterial pressure(MAP) were measured before ischemia,30 min after ischemia,and 30 min,60 min,90 min and 120 min after reperfusion.Myocardial infarction size was determined by TTC staining. Results Baicalin derivant significantly attenuated the declines of LVSP,+dp/dt max,-dp/dt max,HR,SBP,DBP,and MAP.Compared to model group,myocardial infarction sizes were significantly reduced in different Baicalin derivant treated groups [(33.1±6.0)% vs.(29.9±5.4),(24.8±5.0) and(22.3±3.4)%,all P0.05]. Conclusions Baicalin derivant has significant protective effects on myocardial ischemia reperfusion injury in rats.
