摘 要:目的:观察新冠心苏合滴丸对实验性心肌缺血的保护作用。方法:①用异丙肾上腺素复制大鼠急性心肌缺血模型,观察新冠心苏合滴丸对心电图(ECG)的影响,免疫组化检测心脏组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)和一氧化氮(NO)含量。②摘取大鼠心脏,分离并截取相连的左右心房,悬挂、固定,维持在34.5~35.5℃左右;用MS4000U-1C生物信号定量记录分析系统采集不同浓度新冠心苏合滴丸对离体心房肌张力和心搏频率的影响。结果:①新冠心苏合滴丸能明显改善大鼠ECG;提高大鼠心肌组织中NO含量、SOD活性;降低MDA含量,与模型组比有统计学意义(P<0.05,P<0.01)。②有降低离体心房肌张力和减慢心房心搏频率作用;其中200~250mg/L浓度范围作用明显(P<0.05,P<0.01)。结论:新冠心苏合滴丸对大鼠离体心房心肌张力和心搏频率有明显降低作用,对实验性急性心肌缺血模型有一定的保护作用。Objective:To observe the protection of New Guanxin Snhe Dripping Pill (NGSDP)on experimental myocardial is- chemia. Methods :(1)The rats model of myocardial ischemia was established with isoprenaline. The Electrocardiogram(ECG) of rats were determined. Immunohistochemical method was used to detect the activity of superoxide dismutase (SOD) and the content of malondialdehyde(MDA) and nitric oxide(NO) in heart tissue. (2)The heart of rats were taken off,separated and intercepted the a- trium connected around, suspensed and fixed. Temperature maintained at 34. 5-35.5℃. MS4000U-IC biological signal quantita- tive records were used for system analysis to observe the influenee in vitro atrial muscle tension and heartbeat frequency of differ- ent concentrations of NGSDP. Results:(1)NGSDP can obviously improve the ECG of rats;increase the content of NO and the activi- ty of SOD in myocardial tissue ; reduce MDA content, compared with myocardial ischemia model group, NGSDP group was statistically significant( P 〈 0. 05, P 〈 0. 01 ). (2)NGSDP can reduce in vitro muscle tone and slow atrial heartbeat frequency;in the con-centration range of 200 - 250mg/L was obviously( P 〈 0. 05, P 〈 0. 01 ). Conclusion. NGSDP can significant reduce in vitro muscle tone and atrial heartbeat frequency;the effect of NGSDP on experimental myocardial ischemia was protective.
