摘  要:目的研究血小板生成素(TPO)对人巨核细胞白血病细胞(DAMI)增殖、分化的影响。方法采用体外培养巨核细胞,将不同浓度TPO加入细胞培养液,CCK-8法检测TPO最佳刺激浓度;培养液中加入TPO,台盼蓝检测细胞增殖活力,巨核细胞乙酰胆碱脂酶染色检测细胞集落。流式细胞仪检测巨核细胞CD4l表达。结果 TPO浓度为250ng/ml时刺激作用明显。培养液中加入TPO后,巨核细胞集落计数、CD4l表达均明显增加。结论 TPO促进体外巨核细胞增殖、分化。Objective To study the effects of thrombopoietin(TPO) on the proliferation and differentiation of megakaryoblastic leukemia ceils of Human(DAMI). Methods Culture DAMI megakaryocyte in vitro, then added different concentrations of TPO to cell culture system, and detected the best stimulus concentration of TPO by CCK-8). Adding TPO to culture Systems, then detected cell proliferation activity by Trypan Blue, megakaryocytic colony by Acetylcholinesterase staining and the expression of CD41 by flow cytometry in order to observe the effect of TPO on DAMI. Results The stimulation is obvious when the concentration of TPO is 250ng/ml. The megakaryocytic colony and the expression of CD41 are obviously increasing after TPO is added into training liquid. Conclusion TPO can promote proliferation and differentiation of megakaryocyte in vitro.
