摘 要:目的探讨促红细胞生成素(EPO)对大鼠急性下肢缺血性疾病的保护作用。方法将急性下肢缺血性疾病大鼠模型分为生理盐水组和EPO组,按缺血时间不同分为6 h、1 d、2 d、3 d组,采用RT-PCR方法观察HIF-1α变化规律,免疫组化法检测MVD的计数情况。结果通过各时间点比较,EPO组HIF-1α均低于生理盐水组,而MVD均高于生理盐水组,差异有统计学意义(P<0.05)。结论早期应用EPO能明显降低急性下肢缺血大鼠的HIF-1α、提高MVD,对治疗急性下肢缺血性疾病具有重要意义。Objective To observe the the effects of erythropoietin on the expression of hypoxia inducible factor-lot and microvascular density in acute limb ischemia as well as investigate the protection effect of erythropoietin. Methods The ex- perimental rats with acute limb ischemia are separated into two groups: one was injected with 1000 U/kg EPO and the other was injected with 1000 U/kg normal saline. Each group was diyided into 4 subunits by the time of ischemia: 6 hours, 1 day, 2 days and 3 days. The changes of HIF-1α intensity were observed through RT-PCR and MVD counting state was tested through immunohistochemistry. Results The concentration of HIF-1α in rats injected with EPO was lower than the ones injected with normal saline in every subunit and the difference was notable (P 〈 0.05). MVD counting state in rats injected with EPO was higher than the ones injected with NaCL and the difference was notable(P 〈 0.05). Conclusion In the pro- cess of treating acute limb ischemia, injection of EPO in the first few days has obvious therapeutical effect on experimental rats through reducing HIF-1α and improving MVD.
