摘 要:[目的]探讨轮叶党参中分离到的齐墩果酸(OA)对紫外线照射所诱发DNA损伤的保护作用.[方法]取大鼠采用给脾细胞行紫外线照射的方法制作DNA损伤模型,分别给予10,20,40,80mg/L的OA;给DNA损伤大鼠脾细胞加入40mg/L齐墩果酸,培养30,60,90,120min,用荧光分光光度法检测DNA裂解率.[结果]给大鼠脾细胞行紫外线照射后,DNA裂解率明显增高(P<0.01),当OA终质量浓度分别为10,20,40,80mg/L时,紫外线照射之前用OA预处理2h可降低紫外线照射所致的DNA裂解率,DNA裂解率随OA加入量的增加而逐渐降低,当OA剂量超过20mg/L后DNA裂解率下降缓慢.单纯紫外线照射组和加入OA组的DNA裂解率均随着培养时间的延长而逐渐下降,当培养时间达到60min后,两组DNA裂解率间差异有统计学意义,加入OA组的DNA裂解率明显低于单纯紫外线照射组.[结论]齐墩果酸可保护紫外线照射所诱发的DNA损伤,且促进断裂DNA的修复过程.OBJECTIVE To study the protective effects of oleanolic acid(OA) isolated from Codonopsis lanceolata for DNA damage induced by ultraviolet(UV) ray.METHODS UV damage models were established with UV radiated rat splenocytes,and were treated with 10,20,40 and 80 mg/L of OA,respectively.UV radiated splenocytes were treated with 40 mg/L of OA,and then incubated for 30,60,90 and 120 min.DNA lysis rates were detected with fluorescence spectrophotometric method.RESULTS Splenocyte DNA lysis rates were significantly increased after UV radiation(P0.01).At the splenocytes of before UV radiation were pretreated with 10,20,40,80 mg/L of OA for 2 hour,DNA lysis rates were significantly reduced with the dose-dependent.When dose of OA was more than 20 mg/L,the DNA lysis rate was decreased slowly.DNA lysis rates in group of UV radiation alone and group of OA treatment were gradually decreased with extending time of incubating.When the incubating time was 60 min,the difference of DNA lysis rates between both groups had statistically significant,DNA lysis rate of OA treated group was significantly lower than that of pure UV radiation group.CONCLUSION OA might protect DNA from damage induced by UV ray and promote DNA damage repair.
