摘 要:目的 探讨通络方剂(TL)对实验性糖尿病(DM)大鼠肾脏、心脏和腹主动脉组织血管紧张素I(Ang I)和血管紧张素Ⅱ(Ang II)水平的影响.方法 将大鼠随机分为正常对照组(NC组,6只)和DM组未治疗(DM组,6只)、DM+褪黑素治疗组(DM+ML组,6只)和DM+TL组(6只),共治疗8周,以放免法测定肾脏、心脏、腹主动脉组织Ang I、Ang II水平.结果 DM 各组大鼠肾脏、心脏和腹主动脉组织匀浆Ang I、Ang II水平均较NC组明显升高(均P< 0.01);DM+TL组干预后肾脏、腹主动脉组织Ang I、Ang II水平均较DM组显著下降(均P<0.01),与DM+ML组的差异无统计学意义(均P >0.05);DM+TL组和DM+ML组治疗后心脏组织Ang I、Ang II水平与DM组的差异均无统计学意义(均P >0.05).结论 TL能不同程度降低实验性DM大鼠肾脏和腹主动脉组织Ang I、Ang II水平,与ML治疗效果相近.Objective To observe the effects of Tongluo on local tissue angiotensin I (Ang I)and angiotensin II (Ang II) level in kidney ,heart and abdominal aorta in experimental diabetic rats. Methods Male SD rats were randomly divided into 2 group s : normal control group(NC rats,n=6) and diabetic group .Diabetes was induced by STZ (60 mg/kg) abdominal injection. One weeks later, diabetic rats were further divided into 3 groups : diabetic untreated control group (DM rats,n=6), diabetic rats treated with Melatonin (DM+ML rats n=6, 5mg/kg-d) and diabetic rats treated with TongLuo (DM+TL rats, n=6, 1.0g/kg-d). Ra- dioimmunoassay was used to measure tissue(kidney ,heart and abdominal aorta) angiotensin I and angiotensin II level after treat- ed with TongLuo for eight weeks. Results After eight weeks, local tissue angiotensin I (Ang I) and angiotensin II (Ang II ) level in kidney ,heart and abdominal aorta in diabetic untreated group (DM rats) were significantly higher than those in normal rats(NC rats ).Tissue angiotensin I (Ang I)and angiotensin II (Ang II)level in kidney and abdominal aorta in diabetic treated group with TongLuo( DM+TL rats )were significantly lower than those in diabetic untreated group( DM rats )(respectively P〈0.001, P〈0.001, P〈0.001, P〈0.05),but this significant variation was not seen in heart. Conclusion TongLuo administration can decrease the levels of Ang I and Ang II in diabetic kidneys, heart and abdominal aorta in various degree, so it maybe be beneficial to protect against cardiovascular and renal disease progression in diabetes.
