子痫前期和子痫患者胎盘和血浆PDGF-A表达及意义

摘　　要：目的比较正常妊娠和子痫前期、子痫患者胎盘组织血小板源性生长因子(PDGF)-A mRNA、PDGF-AA的表达和血浆PDGF-AA蛋白水平,探讨PDGF-A与子痫前期发病的关系。方法对子痫前期和子痫患者43例、正常妊娠20例,分别用RT-PCR检测胎盘绒毛组织PDGF-A mRNA表达水平,用免疫组化SABC法结合计算机显微图像分析软件,检测胎盘绒毛组织PDGF-AA蛋白的表达水平,用ELISA检测血浆中PDGF-AA蛋白表达水平。结果子痫前期、子痫组及正常妊娠组胎盘组织的PDGF-A mRNA和PDGF-AA蛋白表达,病例组均高于正常组,且与病情轻重呈正相关;病例组血清PDGF-AA蛋白水平均高于正常组,也与病情轻重呈正相关。结论子痫前期和子痫患者胎盘组织及血浆高度表达的PDGF-A可促进动脉粥样硬化形成,可能在子痫前期患者的胎盘血管病变中起重要作用,并参与子痫前期的发生和发展。Objective To compare the expression of platelet-derived growth factor（PDGF）-A mRNA and PDGF-AA in placentas and serum PDGF-AA levels from normal pregnancy,preeclampsia or eclampsia patients,and explore the relationship between PDGF-A and eclampsia.Methods Totally 43 patients with preeclampsia or eclampsia and 20 normal pregnant women were investigated.The expression levels of PDGF-A mRNA and PDGF-AA were respectively determined by RT-PCR and immunohistochemistry（SABC） combined with the analysis of computer microphotography in the placentas.Serum levels of PDGF-AA were determined by ELISA method.Results The expression levels of PDGF-A mRNA and PDGF-AA in placentas from patients with preeclampsia or eclampsia were significantly higher than those in normal group respectively,and showed positive correlation with symptoms of the disease.The serum levels of PDGF-AA in patients were significantly higher than those in normal group and were positive correlation with symptoms of disease.Conclusions PDGF-A highly expression in the placentas of patients with preeclampsia or eclampsia may play an important role in pathological changes of placental vessels in atherogenesis,and is related with the development and progression of the disease.
