摘  要:目的探讨2型糖尿病(T2DM)治疗前后外周血单个核细胞中IL-23p19 mRNA水平以及血浆中IL-23水平的变化。方法检测空腹血糖水平选定分组对象,采用逆转录-聚合酶链式反应(RT-PCR)法检测58例2型糖尿病患者(初诊糖尿病患者28例,经治疗患者30例)和26例健康体检者(对照组)全血中IL-23p19 mRNA的水平,同时采用酶联免疫吸附试验(ELISA)检测各组血浆中IL-23的水平。结果初诊糖尿病组IL-23 mRNA表达水平高于经治疗组和对照组,差异具有统计学意义(P<0.05),IL-23血浆水平比较初诊糖尿病组与经治疗组及对照组差异有统计学意义(P<0.05)。结论 2型糖尿病治疗前IL-23及p19 mRNA水平明显高于经治疗组和对照组,IL-23可能与2型糖尿病的发病和病程进展有一定的关系。Objective To investigate the expressions of interleukin-23p19 mRNA in peripheral blood mononuclear cells(PBMCs) and plasma interleukin-23 level in patients with type 2 diabetes mellitus(T2DM) before and after the therapy.Methods The patients were divided into treatment group(58 T2DM cases included 28 first visitor and 30 treated patients) and control group(26 normal individuals) according to the fasting plasma glucose(FPG).The levels of interleukin-23p19 mRNA in peripheral blood mononuclear cells were measured by reverse transcription-polymerase chain reaction(RT-PCR),and the plasma levels of interleukin-23 were tested by using enzyme-linked immunosorbent assay(ELISA).Results The levels of interleukin-23 mRNA in the treatment group were significantly increased as compared to the control group(P0.05);the plasma levels of interleukin-23 in newly diagnosed T2DM were significantly higher than those in treated patients and control(P0.05).Conclusion The levels of interleukin-23 and p19mRNA were significantly increased in untreated T2DM patients compared with after treatment and control,interleukin-23 may be involved during the occurrence and development of T2DM.
