摘  要:以小鼠为对象,侧脑室内埋植慢性套管,行为训练后icv精氨酸加压素(AVP)1.5 ng/鼠,对抑制性回避反应的记忆保持有增强作用.行为训练前ip肾上腺素β受体阻滞剂Pro 0.5mg/kg,能阻断AVP icv对记忆保持的易化.摘除小鼠双侧肾上腺,不改变外周注射AVP 10μg/kg所产生的促记忆效应.结果提示AVP促记忆作用的部位可能在中枢,并与脑内肾腺β受体有关.We previously reported that peripheral injection(ip)of AVP facilitated memory retention in an inhibitory avoidance(IA)task in mice.The retention facilitation of AVP was blocked by propranolol(Pro).In present study,we investigated the behavioral effect of AVP administered centrally,with an attempt to relate that effect of adrenal β-receptor in the brain.Male Kun-ming mice were used in two set of experiments.They were trained in the IA task.The retention latency was recorded as a measure of memory.In the first experiment,chronic cannulae were implanted into the lateral cerebral ventricle of mice 4-5 d before the behavioral test.Immediately posttraining,several doses of AVP(0.1,0.5 and 1.5ng/mouse)or normal saline(NS)were injected through the cannulae into the cerebral ventricle(icv).Only AVP 1.5 ng/mousegroup had the longer retention latencies in the IA task when compared with the NS group,(p<0.05).Pretraining ip injection of Pro(0.5 mg/kg),i.e.the same dose we used in previous study in the same task,also blocked the effect of AVP(icv).In the second experiment,mice were subjected to adrcnalectomy(ADX)or sham-operation(SHAM).Behavioral test were carried out 7-10 d after surgery.The memory enhancing effect of AVP(10μg/kg ip)still retained in mice both with ADX and SHAM.Above data suggest that the behavioral effect of AVP was probably mediated centrally and adrenal β-receptor in the brain may be involved in the memory facilitation effect of AVP.
