摘 要:目的研究日本大耳白兔胎兔动脉导管组织中是否存在一氧化碳/血红素加氧酶-2(CO/HO-2)体系的表达。方法取日本大耳白兔孕兔4只(每只均受孕,平均每只孕兔产仔兔6只),共产仔兔24只,取各仔兔动脉导管组织和心脏血,RT-PCR和Westernblot分别检测仔兔动脉导管组织中是否有HO-2的表达,实时荧光定量PCR检测仔兔动脉导管组织中HO-2mRNA的表达量;内源性CO试剂盒(全血测定)检测各仔兔新鲜心脏血中内源性CO的含量。结果仔兔动脉导管组织中存在HO-2mRNA及HO-2蛋白的表达;HO-2mRNA表达量为0.33±0.11;仔兔心脏全血中CO的含量为(53.09±6.73)umol/L。结论胎兔动脉导管组织中存在一氧化碳/血红素加氧酶-2(CO/HO-2)体系的表达。Objective To study whether there is the expression of carbon monoxide/heme oxygenase-2(CO/HO-2) system in the tissue of arterial duct in Japanese big-ear fetal rabbits.Methods Japanese big-ear rabbits(n=4) were selected(each was pregnant and every rabbit produced average 6 fetal rabbits),and totally there were 24 fetal rabbits were produced.The samples of tissue of arterial duct and heart blood were collected from all fetal rabbits.Reverse transcription-polymerase chain reaction(RT-PCR) and Western blot technique were used to detect whether or not there is the expression of HO-2 in the tissue of arterial duct.The expression quantity of HO-2 mRNA was detected by applying real-time fluorescence quantitative polymerase chain reaction(FQ-RT-PCR),and content of endogenous CO was detected by using endogenous CO kit(whole blood determination) in fresh blood of fetal rabbits.Results There were the expression of HO-2 mRNA and HO-2 protein in the tissue of arterial duct of fetal rabbits.The expression quantity of HO-2 mRNA was 0.33±0.11.The results of endogenous CO kit showed that CO content in whole heart blood was(53.09±6.73) umol/L.Conclusion There are some expression of CO/HO-2 system in the tissue of arterial duct.
