摘 要:目的:观察五苓散对腹泻模型大鼠结肠AQP4及AQP4mRNA表达的作用,揭示五苓散止泻作用的机理。方法:将SD大鼠随机分为正常对照组、腹泻模型组、氢氯噻嗪组、口服盐液组、五苓散低、中、高剂量组7组,观察大鼠腹泻状态并测体重等。分别于第4天上午、第7天上午取大鼠结肠,免疫组化法检测AQP4蛋白的表达,原位杂交方法检测AQP4 mRNA的表达。结果:腹泻模型组、口服盐液组、五苓散低剂量组结肠AQP4及AQP4mRNA表达明显下调,氢氯噻嗪组、五苓散中剂量组AQP4及AQP4mRNA表达下调但明显高于模型组,五苓散高剂量组结肠AQP4及AQP4mRNA表达下调不明显。结论:AQP4及AQP4mRNA表达下调是番泻叶致大鼠腹泻的机制之一,上调腹泻模型大鼠结肠AQP4及AQP4mRNA表达是五苓散的止泻机理之一,但与剂量相关,5.4g/kg灌胃效果较好。Objective : To study the effect of Wu Ling San-prescription of traditional Chinese medical -on expression of AQP4 and AQP4mRNA in colon mucosa of rat model of diarrhea induced by senna leaf. Methods: SD rats were randomly divided into 7 groups: normal control group, model of diarrhea group, Hydrochlorothiaxide group, oral rehydration salts (ORS) group, Wu Ling San low-dose, moderate-dose, high-dose group. Except normal control group, all rats were given by gastrogavage once a day in every day morning with senna leaf liquid (20ml/kg, 3.6g/kg) to establish rat model of diarrhea and different treat were given in the afternoon. After observing the rat' s general situation, faces appearance and measuring the weight every day, we take the colon at 5cm away from ileocecal orific from each half number of rats in every group at 4d and 7d. Immunohistochemical technique was used to test the expression of AQP4 in the colon mucosa. In situ hybridization was used to test the expression of AQP4mRNA in the colon mucosa. Results: Compared with the normal control group, expression of AQP4 and AQP4mRNA were decreased significantly in colon mucosa of rat model of diarrhea group, ORS group and Wu Ling San low-dose group at 4d, 7d. Expression of AQP4 and AQP4mRNA were decreased in colon mucosa of rat in Wu Ling San moderate-dose group at 4d, 7d, but higher than model of diarrhea group. Expression of AQP4 and AQP4mRNA in colon mucosa of Wu Ling San high-dose group found no significant decreased compared with normal control group and higher than model of diarrhea group. Conclusion: Down regulated of expression of AQP4 and AQP4mRNA in colon mucosa of rat is one of a mechanism of action of senna leaf to make rat diarrhea. Up regulated expression of AQP4 and AQP4mRNA in colon mucosa of diarrhea rat is one of a mechanism of action of Wu Ling San to stop diarrhea. But being dose-response relationship and each day 5.4g/kg is the best dose.
