摘  要:目的:确定沼泽红假单胞菌培养的最优化培养基成分组合和培养条件,为沼泽红假单胞菌直接作为光合叶面肥应用提供试验数据.方法:采用单因素试验设计分别对沼泽红假单胞菌培养的接种量、pH值、温度、光照强度四种因素的不同水平进行比较试验,确定其各自最佳试验水平;采用L9(34)正交试验设计从9种培养基成分组合中选择出最优化试验配方组合.结果:沼泽红假单胞菌生长最优条件为:接种量为15%、pH值为7.2、温度为35℃、光照强度为2000xl时;最佳培养基组成配方为:NH4Cl1.0g、K2HPO40.5g、NaHCO33.0g、酵母膏2.0g.结论:优化的生长条件和培养基配方组成能有效促进沼泽红假单胞菌生长和繁殖.Objective: The optimization of culturing conditions and medium component for Rhodopseudomonas palustris is studied, in order to obtain the experimental data of the application of PSB that uses directly the surface of leaf as a bacterial fertilizer. Method: To determine respectively the optimal level of factors in the growth conditions of Rhodopseudomonas palustris, including the inoculum size, pH value, temperature and light intensity, with single factor experiment design, and to use L9(34) orthogonal layout of the orthogonal design to test the optimization ingredients on the culturing medium components. Results: The optimal growth conditions of Rhodopseudomonas palustris are an inoculum size, 15%; pH value, 7.2; temperature, 3℃; and light intensity, 20001x. The best medium ingredients are ammonium chloride 1.0g, hydrogen phosphate potassium 0.5g, sodium bicarbonate3.0g and yeast extract 2.0g. Conclusion: The optimized culture medium ingredient and the best level combination of growth conditions can effectively improve Rhodopseudomonas palustris growth and propagation.
