摘 要:目的探讨黑头发和白头发线粒体DNA(mtDNA)控制区的异质性差异。方法提取毛干mtDNA,用PCR技术扩增mtDNA控制区5个目标片段,以变性高效液相色谱技术分析中国汉族正常人与Alzheimer病同个体黑头发和白头发mtDNA控制区目标片段的异质性差异。结果 30~40岁正常人、50~70岁正常人和50~70岁Alzheimer病患者黑头发mtDNA异质性分别为:3.2%、10.0%及16.4%;30~40岁正常人、50~70岁正常人和50~70岁Alzheimer病患者白头发mtDNA异质性分别为:7.2%、20.4%及28.0%,各年龄段正常人及Alzheimer病患者同个体白头mtDNA异质性明显高于黑头发。结论同个体中黑白头发和mtDNA控制区的异质性有明显差异,年龄越大,异质性越明显。Objective To investigate the heterogeneity differences in mtDNA control region between black and white hairs.Methods The mtDNA was extracted from hair shaft from black and white hairs of health participants and patients with Alzheimer′s disease(AD) in Chinese Han population.PCR and DHPLC were applied to amplify five target DNA fragment of mtDNA control region and analysis of heterogeneity differences in mtDNA control region between black and white hairs from single subject.Results The mtDNA heterogeneities in black hair from normal controls of 30~40 and 50~70 years old,and AD patients of 50~70 years old were 3.2%,10% and 16.4%,respectively;while in white hair were: 7.2%,20.4%,28%,respectively.The mtDNA heterogeneities in white hairs were significantly higher than those in black hairs.Conclusion There are significantly differences in heterogeneity of mtDNA control region between black and white hairs in single individuals,with more prominent in older population.
