摘 要:目的研究稳恒磁场对体外培养的人鼻咽癌细胞(CNE-2)生长抑制与凋亡作用的影响。方法利用磁感应强度为7 mT的自制稳恒磁场对体外培养的CNE-2细胞进行照射,经1、3、9、12、24、36、48、60、72 h不同时间照射处理后,采用台盼蓝拒染法进行活细胞计数,MTT法检测细胞生长活性,HE染色观察细胞形态学改变,琼脂糖凝胶电泳观察细胞凋亡情况。结果从照射9 h起细胞生长受到显著抑制(P<0.05),生长曲线结果与MTT实验具有良好的一致性。经72 h照射处理后,MTT法、HE染色观察细胞出现形态学改变,但琼脂糖凝胶电泳检测未见细胞DNA断裂现象。结论 7 mT稳恒磁场照射对体外培养的CNE-2细胞生长有显著的抑制作用,而对诱导细胞凋亡的效果有待进一步研究。Objective To study the influence of steady magnetic field on the growth and apoptosis of human nasopharyngeal carcinoma cells CNE-2.Methods CNE-2 cells cultured in vitro were exposured by the self-made steady magnetic field with the magnetic induction intensity of 7 mT,after 1,3,9,12,24,36,48,60,72 h of insolation magnetism processing,Trypan blue exclusion was used to determine the viable count,MTT was used to detect the growth rate of cells;the morphologic change of cells was observed by HE,DNA degradation was detected by the agarose gel electrophoresis.Results The remarkable depressive effect of magnetic field for the cell growth starting from 9 h(P〈0.05),the growth curve of CNE-2 cells had good consistency with MTT result.After 72 h insolation magnetism processing,the cells showed morphologic changes,but had no DNA breakage.Conclusions 7 mT steady magnetic fields have significant depressive effect to CNE-2,but the effect of inducing cell apoptosis need further study.
