摘  要:目的观察毒胡萝卜素对ReNcell CX人神经干细胞存活、增殖和凋亡的影响。方法将ReNcell CX人神经干细胞分为两组,实验组分别加入10、20、50、100 nM毒胡萝卜素,干预6 h;对照组不加任何药物。采用分光光度计法测量两组乳酸脱氢酶(LDH)水平,免疫荧光显微镜下观测细胞增殖情况及Caspase-3水平。结果实验组LDH、Caspase-3水平明显高于对照组,细胞增殖率明显低于对照组,且均呈剂量依赖性,P<0.05或<0.01。结论毒胡萝卜素抑制神经干细胞存活和增殖;其机制可能为激活Caspase-3释放,促进神经干细胞凋亡。Objective To investigate the influences of thapsigargin on the survival, proliferation and apoptosis of hu- man neural stem cells. Methods The human neural stem cells ReNcell CX were randomly divided into two groups, the experimental group was treated with thapsigargin of different concentrations( 10, 20, 50 and 100 nM respectively) ; the control group was given no drugs. The LDH release was measured via Spectrophotometer method, the cell proliferation and Caspase-3 level were obtained by immunofiuorescent stain. Results The LDH and Caspase-3 level in the experimental group were significantly higher than those in the control group, thapsigargin inhibited the proliferation and induced the Caspase-3 release of the ReNcell CX stem cells in dose-dependent manner(P 〈 0.05, 0.01 ). Conclusions Thapsigargin can inhibit the survival and proliferation of ReNcell CX human neural stem cells, which may be caused by promotion of ReNeell CX human neural stem cells anoDtosis via activation of CasDase-3.
