结直肠癌组织中肝细胞生长因子表达与微血管计数的相关性研究

摘　　要：为探讨肝细胞生长因子（HGF）在人结直肠癌组织中的表达与肿瘤组织微血管生成之间的关系及其临床意义，本研究采用免疫组织化学SP法对65例人结直肠癌组织及其癌旁组织标本进行HGF检测，用CD34抗体标记微血管计算微血管计数（MVC），并分析HGF高表达和MVC与淋巴结转移、肝转移、TNM分期的相关性。结果显示，HGF在结直肠癌组织中的高表达（＋＋、＋＋＋）率为83．08％（54／65），癌旁组织中为12．31％（8／65），两者差异有统计学意义，P〈0．01；结直肠癌组织中HGF表达及MVC与有无淋巴结转移、肝转移及TNM分期有关，且淋巴结转移、肝转移、T3,4期患者结直肠癌组织中HGF高表达率及MVC显著高于无淋巴结转移、无肝转移及T1,2期结直肠癌患者，P〈0．05；HGF高表达、MVC与癌转移显著相关，高血管组（MVC≥16）HGF高表达及肝转移例数明显多于低血管组（MVC〈16），P〈0．01。结果表明，HGF的异常表达与人结直肠癌血管生成密切相关，其表达可作为判断结直肠癌侵袭、转移的指标。In order to explore the expression of hepatocyte growth factor （HGF） in human colorectal canc- er tissues, and its relationship with micro-vessel formation in neoplasms tissues, as well as clinical signifi- cance,this study adopted immunohistochemistry SP method to detect the HGF included in the human colo- rectal cancer tissue and its around-cancer tissue samples of 65 cases,calculated micro-vessel counts （MVC） by using CD34 antibody labelling,and analysed the correlation of high expression of HGF, and MVC with lymph node metastasis,liver metastasis,and TNM staging. As results,the high expression rate of HGF in colorectal cancer tissues was 83.08%（54/65, ＋＋, ＋＋＋）, was 12.31%（8/65, ＋＋, ＋＋＋） in around-cancer tissues, there was statistically significant difference（ P 〈0.01） ;the expression of HGF in colorectal cancer tissues and MVC were related to lymph node metastasis,liver metastasis and TNM staging, and the high expression rate of HGF in colorectal tissues and MVC in the patients with lymph node metastasis,liver metasta- sis, T3,4 stage were significantly higher than that in the patients without lymph node metastasis and liver metastasis as well as being T1.2 stage（ P 〈0.05）;high expression of HGF, MVC and cancer metastasis were of high correlation;in high vessel counts group （MVC≥16） ,high expression of HGF and case number of patients with liver metastasis were significantly more than that in low vessel counts group （MVC% 16）, P 〈0.01. Results show that the abnormal expression of HGF is closely related to the micro-vessel formation in human colorectal cancer tissues,thus the expression could be served as the indexeS for judging colorectal cancer invasion and metastasis.
