摘  要:应用低速离心和巨细胞病毒早期核抗原单克隆抗体间接免疫荧光方法(简称DEACF),检测临床标本中巨细胞病毒(CMV),并与传统病毒分离方法进行比较。结果:139份临床标本病毒分离方法阳性16份(11.5%),细胞病变(CPE)出现时间平均9.2日,其中2份DEACF检测阳性;DEACF检测阳性18份(13.0%),培养16h61.1%出现阳性,培养88h可检出全部阳性标本,其中4份病毒分离阴性标本中,3份有证据不是DEACF的假阳性反应。DEACF的敏感性为89.5%,特异性为99.2%。DEACF检测的139份标本中,用低速离心可使培养88h的阳性检出率从未离心的44.4%提高到100%。A monoclonal antibody was used to detect an early nuclear antigen of CMVfrom clinical specimens which were inoculated onto human embryo lung cellswith low centrifugation and indirect immunofluorescence (DEACF). 139 speci-mens were processed in both standard tube cell culture and 24 well cell cultureplate. Among them 16 (11.5%) were positive in standard tube cell culture (theappearance of CPE average 9.2 days) and 18 (13%) were positive in 24 wellcell culture plate at 88 hr postinfection with DEACF. 4 specimens werepositive by DEACF but negative by virus isolation, 3 specimens had evidenceof not false-positive. The sensitivity and specificity of DEACF is 89. 5% (17/19)and 99.2 % (119/120), respectively. 139 specimens were examined by DEACF.The positive rate increased from 44.4 % without low centrifugation to 100%with low centrifugation at 88 hr postinfection. There was significant difference.It is suggested that the infectivity of CMV can be enhanced by low centrifu-gation, so the time of detecting CMV from clinical specimens is shortened. Incomparing DEACF with virus isolation, it is suggested that DEACF is a rapidsensitive and specific method in diagnosis of CMV infection and can be widelyused in clinical virus laboratories.
