当归补血汤对移植肌卫星细胞γ射线照射小鼠早期造血重建作用的影响

摘　　要：目的观察当归补血汤对移植骨骼肌卫星细胞的受体小鼠早期造血功能重建的影响。方法分离培养雄性同系小鼠的肌卫星细胞并鉴定。经8Gy137Cs-γ射线照射的雌性昆明种受体鼠随机分为6组:空白对照组、移植肌卫星细胞组、当归补血汤不同剂量(1,3,5,10倍灌胃7 d)干预的移植肌卫星细胞4组。观察移植后8周内受体鼠存活率、外周血血常规、脾脏指数、脾形态学的变化,并采用PCR方法鉴定其造血重建的来源。结果培养的肌卫星细胞呈desmin染色阳性;当归补血汤各剂量组受体鼠脾脏指数、脾结节数、白细胞数、血小板数、血红蛋白数、8周内存活数均高于空白对照组和移植肌卫星细胞组,特别是给药3组及4组(P<0.05);对移植的存活小鼠进行Y染色体PCR分析,证实重建雌性受体鼠造血功能的细胞来自雄性供体。结论经当归补血汤干预后,肌卫星细胞能使受体鼠早期造血功能得以重建。Objective To investigate the effect of Danggui Buxue Decoction on the early period hematopoietic reconstruction of mice transplanted by muscle satellite cells.Methods MSC were procured from new born male mice.The desmin protein of the cultured MSC was examined by immunohistochemistry.The female mice irradiated with 8 Gy 137Cs-γ ray were randomly divided into 6 groups：control group,the transplanted MSC group and four groups disposed by the different doses of Danggui Buxue Decoction after transplantation.The changes of survival rate,peripheral blood routine examination,spleen index,spleen morphological changes were observed.PCR was used to verify the cell source of hematopoietic reconstruction.Results The desmin protein was positive in the cultured MSC.The number of CFU-S,RBC,platelet,HGB and 8 week survival mice in Danggui Buxue Decoction group were higher than those of control group.Especially group 3 and group 4 disposed by Danggui Buxue Decoction were significantly higher than the normal control or the transplanted MSC group（P0.05）.PCR results showed Y chromosome sequence existed in the survived mice of each transplanted cell group,indicating that the repopulated hematopoietic cells came from the male donor.Conclusion The transplanted cell can reconstruct the hematopoiesis in mice better and earlier under the influence of Danggui buxue Decoction.
