摘  要:目的建立多西他赛长循环脂质体包封率的测定方法。方法以高效液相色谱法测定,考察透析介质的种类、浓度、透析袋内外体积比、温度等对脂质体与游离药物分离效果的影响。结果透析介质为0.5%聚山梨醇酯,透析袋内外体积比为1∶150,在0℃下透析8 h达到平衡。结论透析法准确可行,适于多西他赛长循环脂质体的包封率测定。Objective To establish a method for determination of the entrapment eiticiency ot long - clculatlng llposomes entrapped with docetaxel( DOC - PEG2000 - DSPE - Liposomes, DPL). Methods HPLC was used to determine and investigate the entrapment efficiency of DPL the sort of the release media, concentration, and the volume ratio between internal and external. Results The release media was 0.5% polysorbate, the volume ratio between internal and external was 1:150, the equilibrium time was 8 h. Conclusion Dialysis method is accurate. It is suitable for determination of entrapment effiency of DPL.
