摘  要:目的研究乌头炮制过程中生物碱变化对实验大鼠毒性作用的影响。方法通过大鼠毒性实验,探讨炮制过程中生物碱的变化对心、肝、肾脏的毒性作用差异及对心脏损伤作用的机制。结果从生品中提取得到的双酯型生物碱(DDA)毒性较大,对心、肝、肾脏损伤较重,生物碱对心脏损伤的机制与脂质过氧化损伤和细胞凋亡有关。结论乌头炮制过程中生物碱的变化降低了对实验大鼠心、肝、肾脏的毒性,其机制与生物碱变化影响了凋亡相关蛋白Caspase-3、Bcl-2蛋白的表达及减少心肌细胞内的氧化损伤有关。Objective To study the changes of alkaloids by processing in Aconirum. L and its toxicity. Methods The toxic experiment was adopted to investigate the changes of alkaloids in Aconirum. L induced by processing on cardiac muscle damage and apoptosis. Results The model of cumulative intoxication showed that different alkaloids extracted from different aconite roots and processing products had the different degree toxicity in biochemistry, organization, cell, deuto - cell and molecular level. DDA could decrease the activity of SOD and increase the level of MDA compared with MDAH and MDAP groups. The mechanism of toxic effect of DDA was related with occurrence of apoptosis and excessive expression of the protein caspase - 3 and lower expression of bcl - 2 protein. Conelusion The mechanism of attenuation in processing is related to the changes of alkaloids, which decrease the injury of cardiac mitochondrome and apoptosis by regulating the expression of caspase - 3 and Bcl - 2 protein.
