摘  要:目的滨蒿内酯(Soparone,Sco.)通过松弛气道平滑肌、清除气道氧自由基、抑制气道炎症等多种途径起到抗炎平喘作用,该实验基于前期所做的研究工作,进一步探讨了Sco对肥大细胞(mast cells,MCs)脱颗粒及其炎症介质的释放作用的影响及可能的调控途径。方法不同浓度的Sco(10-5、10-6和10-7mol/L)分别作用于正常及致敏MCs,并设立正常及致敏MCs的溶剂对照组(control,asthma),比较各组MCs脱颗粒百分数、类胰蛋白酶活力、组胺释放率及MCs细胞内钙([Ca2+])i的变化。结果 1×10-5和1×10-6mol/L Sco.组MCs脱颗粒百分数分别为(39.10±2.81)%和(11.40±2.37)%,明显低于asthma组的(78.90±4.12)%(P<0.05),且以上Sco.组MCs的组胺和类胰蛋白酶释放亦较asthma组显著减少(P<0.05);在无细胞外钙条件下,不同浓度的Sco(10-5、10-6和10-7mol/L)对正常MCs[Ca2+]i的影响分别为(-12.1±2.2)%、(-6.1±2.4)%和(-2.1±0.7)%,对致敏MCs[Ca2+]i的影响分别为(-75.6±2.2)%、(-33.9±2.5)%和(-4.0±0.02)%,相同浓度Sco.对正常组和致敏组MCs[Ca2+]i的降低作用的组间差异有显著性(P<0.05)。结论 Sco.可剂量依赖性抑制豚鼠MCs脱颗粒从而抑制MCs组胺及类胰蛋白酶的释放;Sco.能够抑制豚鼠正常MCs以及致敏MCs[Ca2+]i水平。[ Objective ] In our previous studies we have confirmed scoparone(Sco.) can relax airways smooth mus- cle in guinea pig. The aim of study was to investigate the effect of scoparone on mast cells (MCs) releasing histamine and tryptase and regulating intracellular calcium in guinea pig, to provide theory foundation about asthmatic cure in clinic. [Methods ] Different concentrations of Sco.(1×10-5 mol/L, 1×10 mol/L, 1×10-7 mol/L) were added to sensi- tized MCs in vitro, and then degranulation of MCs, histamine releasing of MCs and tryptase activity of MCs were determined by microscopy, spectrofluorometer and UV spectrophotometer, respectively. In single MCs, [Ca2+]i was measured by intracellular imaging fluorescence microscopy system. [ Results ] In this study, we found that different concentrations of Sco.(1×10-5 mol/L, 1 ×10-6 mol/L) could affect peritoneal MCs degranulation, comparing to asthma group, the phenomenon of MCs degranulation were significantly inhibited(P 〈0.05); Under the circumstances of MCs sensitized in vitro, compared to control group, asthma group was much higher than control group on releasing his- tamine and tryptase (P 〈0.05) and different concentrations of sco. group (1×10-5 mol/L, 1×106 tool/L) compared to asthma group, the releasing of histamine and tryptase were depressed (P 〈0.05); In normal and sensitized MCs of guinea pig, different concentrations of Sco. group (1×10-5mol/L, 1×10-6 mol/L, 1×10-7 mol/L) could significantly dose- dependent inhibit MCs intracellular calcium release compared to asthma group (P 〈0.05). [ Conclusions ] Sco. could dose-dependent inhibit MCs releasing histamine and tryptase and decrease [Ca2+]i of normal and sensitized MCs in guinea pig.
