金钱树四倍体的诱导与鉴定

摘  要:以金钱树块茎为材料,采用秋水仙素溶液处理不定芽生长点,研究秋水仙素浓度和处理时间的诱变效果。结果表明:0.1%的秋水仙素溶液浸泡处理块茎的不定芽48 h效果最好,其诱导率为30%。通过流式细胞仪对金钱树植株DNA相对含量的测定,鉴别出四倍体和嵌合体。四倍体植株与二倍体植株在叶厚、保卫细胞大小、气孔密度等方面存在明显差异,其中气孔的形态及密度可作为鉴别四倍体、嵌合体和二倍体的重要性状。Rhizome of Zamioculcas zamiifolia treated with different concentrations of colchicine solution at different stages in order to figure out the best inducing method and obtain tetraploid. The results showed that the polyploids could be most effectively induced by immersing buds in 0.1% colchicines solution for 48 hours, the inductive frequency was 30%. Ploidy level changes were confirmed via flow cytometry, some belonged to tetraploid plants and others were different mixoploid. Comparing the characteristics of leaves and stomata of tetraploids and diploids, the results showed that these characteristics including thickness of leaf, length and width of guard cell, density of stoma were distinctly different from diploid. The morpha and density of stoma could be used to distinguish diploids, mixoploid and tetraploids.
