加味四君固本汤与Iressa联合抑制肺癌细胞增殖的实验研究

摘　　要：目的探讨自拟加味四君固本汤含药血清与表皮生长因子受体(EGFR)靶向治疗药物Iressa联用对人非小细胞肺癌细胞系A549及HCC827增殖的抑制作用。方法大鼠灌胃制备高、中、低剂量加味四君固本汤含药血清,采用MTT法检测加味四君固本汤含药血清单用以及联合Iressa使用对人非小细胞肺癌细胞系A549和HCC827增殖的影响。结果 Iressa抑制A549和HCC827细胞增殖的半数抑制浓度(IC50)分别为46.18μmol/L和61.99μmol/L(P<0.05);加味四君固本汤含药血清作用于肺癌细胞后,随着含药血清浓度的逐渐增加,抑制率逐渐增高(P<0.05);加味四君固本汤含药血清与Iressa联用加强了对A549和HCC827细胞增殖抑制作用(P<0.05)。结论加味四君固本汤含药血清具有一定的抗肺癌作用,并存在一定的量效关系,联合Iressa使用可协同抑制A549和HCC827细胞增殖。Objective To evaluate the inhibiting effects of self-made Jiawei Sijun Guben Decoction in combination with Iressa on non-small-cell carcinoma lines A549 and HCC827. Methods Rats were given high, medium and low concentrations of Jiawei Sijun Guben Decoction by gavage to prepare drug-containing serum. MTT assay was used to detect the proliferation of non-small-cell carcinoma lines （A549 and HCC827） treated with drug-containing serum in combination with Iressa. Results The ICs0 of Iressa on A549 and HCC827 cell lines was 46.18 ~mol/L and 61.99 ~mol/L. The inhibiting effects on A549 and HCC827 were significantly increased with the increase of drug-containing serum （P 〈 0.05）. Synergistic inhibiting effect appeared when drug-containing serum in combination with Iressa （P〈0.05）. Conclusion Jiawei Sijun Guben Decoction drug-containing serum can inhibit the proliferation of non-small-cell carcinoma lines by the manner of dose-effect. Synergistic inhibiting effects were found when drug-containing serum in combination with Iressa.
