摘  要:目的:观察败血症时肝细胞核Ca2+摄取及释放功能的改变。方法:通过结扎并穿刺盲肠制作大鼠败血症模型,差速离心制备细胞核,进行45Ca2+摄取和释放测定。结果:败血症早期组当介质中游离[Ca2+]为200、400、1000nmol·L-1时,核Ca2+摄取比对照组分别增加35%、95%、160%(P<0.01)。晚期组分别只有对照的87%~88%(P<0.05)。在10μmol·L-1三磷酸肌醇作用下,早期组1min内释出其摄入量的76%,而晚期组仅为52%,与对照组的63%差异有显著性(P<0.01)。结论:败血症时核钙摄取和释放功能呈早期增加,晚期减弱的双向变化。Objective: To investigate the alteration of Ca 2+ uptake in different Ca 2+ concentration and Ca 2+ release by isolated rat liver nuclei during sepsis. Methods: The sepsis was prepared on rat by cecal ligation and puncture. 45 Ca 2+ uptake and release assay were performed on rat liver nuclei isolated by differential centrifugation. Results: The nuclear Ca 2+ uptake in early sepsis group (9 h after operation) increased significantly by 35%, 95% and 160% ( P <0.01) when the freeCa 2+ in medium was 200, 400, 1 000 nmol·L -1 , respectively. While the late sepsis group (18h after operation) was only 87% 88% of the according normal values ( P <0.05). The Ca 2+ release percent was 76 % in the first minute after adding 10 μmol·L -1 inositol 1,4,5 trisphosphate (IP 3) in early sepsis group, while the late sepsis group was only 52 %, both showed significant difference from the normal 63 %( P <0.01). Conclusion: The Ca 2+ uptake and release by isolated rat liver nuclei increased during early sepsis, but decreased during late sepsis.
