摘 要:[目的]观察半夏秫米汤对睡眠剥夺大鼠八臂迷宫空间学习记忆能力的影响。[方法]使用随机平行对照方法,将清洁级Spargue-Dawley雄性成年大鼠40只适应性喂养3d后随机分为正常对照组、模型组、半夏秫米汤高、中低剂量组,每组8只。用D.Suchecki改良多平台法进行睡眠剥夺。同时开始给药,经胃灌服半夏秫米汤(半夏20g,秫米30g)。按人体用药量10倍、5倍、2倍换算用药量,分别为10g/(kg.d)、4g/(kg.d)、2g/(kg.d),灌胃容积为1 mL/kg。其它两组灌服等体积的蒸馏水。于000时进行旷场实验(水平运动、垂直运动)、八臂迷宫实验(行走时间)。观测行为学及记忆能力变化。[结果]旷场实验与正常对照组比较,模型组大鼠的水平运动得分及垂直运动得分明显下降(P<0.01);与模型组比较,中药低、中剂量组大鼠的水平运动得分及垂直运动得分明显上升(P<0.05或P<0.01)。八臂迷宫记忆能力检测与正常对照组比较,模型组所用时间、错误总数升高(P<0.01),首错前正确数降低(P<0.05);与模型组比较,中药低、中剂量组大鼠所用时间、错误总数降低(P<0.05或P<0.01),首错前正确数升高(P<0.05或P<0.01)。[结论]半夏秫米汤具有改善睡眠剥夺大鼠焦虑行为、好奇度降低及空间学习记忆能力的作用。[ Objective ] To explore the effect of Banxia Shumi decoction on on space learning and memory ability in Radial Maze of the rat model of sleep deprivation. [ Methods ] Use the rat model of sleep deprivation as research object, and then randomly divided all rats into five groups and the rats of each group intragastrically perfused with corresponding interventions. After the experiment, the behavioral of open - field test and the behavioral indicators of Radial Maze were examined. [ Results ] The behavioral indicators of open filed test: Compare with blank group, the behavior score of horizontal movement and vertical movement of open - field test were lower in model group ( P 〈 0.01 ) , compared with the model group, the behavior score of horizontal movement and vertical movement of open - field test were lower in model Banxia Shumi decoction lower and middle dose treating group( P 〈0.01). The behavioral indicators of Radial Maze. the time of walk over the Radial Maze, the whole wrong times in model group were higher than them in blank group ( P 〈 0.05 or P 〈 0.01 )and the right times before the first wrong appeared in model group was lower than it in the blank group ( P 〈0.05 orP 〈0.01 ). the whole wrong times in Banxia Shumi(low dose and middle dose) decoction treating group were lower than them in model group( P 〈0.05 or P 〈0.01 )and the right times before the first wrong appeared in Banxia Shumi (low dose and middle dose) decoction treating group was higher than it in model group( P 〈0.05 or P 〈0.01 ). [ Conclusion] Banxia Shumi decoction has the effect of space learning and memory ability in radial maze of the rat model of sleep deprivation.
