摘  要:目的探讨血清总胆汁酸(TBA)作为山豆根致肝损伤早期敏感标志物的可能性。方法取Wistar大鼠,随机分为对照组和山豆根水煎液组(简称山豆根组),山豆根组每天灌胃给予水煎液6 g·kg-1,对照组灌服蒸馏水,于给药1 d、7 d、16 d分批处理动物。测定常规血液生化指标—丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TB)、碱性磷酸酶(ALP)、总蛋白(TP)、白蛋白(ALB)和TBA的水平;计算肝脏指数,并进行HE染色,观察肝组织病理形态学变化。另取正常动物,测定肝、肾、脾、肺、心、脑、胸腺组织中TBA的含量。结果与同时间点对照组相比,山豆根组常规生化指标ALT表现出良好的时效关系,于给药7 d、16 d升高(P<0.01),AST于给药1 d升高(P<0.01),其他时间点无明显变化,肝脏指数和肝组织病理形态学检查无明显变化。TBA于给药1 d、16 d升高(P<0.01),变化较ALT早,与同时间点对照组相比,TBA数值的变化倍数高于ALT和AST。组织含量测定显示肝组织中TBA含量较高。结论 TBA可作为山豆根致肝损伤的早期敏感生物标志物之一。Objective To study the possibility of total bile acid(TBA) as an early and sensitive marker for liver in- jury rats induced by Radix Sophorae Tonkinensis(RST). Methods Wistar rats were divided randomly into control group and RST group. RST group was orally administrated with the decoction from RST at the dose of 6 g·kg^-1 once a day, while the distilled water was given to the control group in the same manner. Rats were sacrificed on day 1, day 7 and day 16 after administration. The activities of serum alanine aminotransferase (ALT) and aspartate aminotrans- ferase (AST), and the levels of total bilirubin(TB), alkaline phosphatase(ALP), total protein (TP), albumin (ALB) and total bile acid(TBA) were determined. The liver was weighed, and the ratio of liver / body weight was calculated. Liver pathological changes were observed by HE staining. The content of TBA in the tissues of liver, kidney, spleen, lung, heart, brain, thymus were determined in normal rats. Results Compared with control groups, ALT activity which had time-effect relationship was significantly increased on day 7 and day 16(P 〈 0.01). AST activity was ele- vated only at day 1 (P 〈 0.01 ). Neither heptatic index nor hepatic pathological features showed obvious changes. TBA was elevated on day 1 and day 16(P 〈 0.01). TBA was demonstrated to be an earlier change. The difference of TBA value was much greater than that of ALT and AST. The content of TBA in liver was higher. Conclusion TBA may serve as a sensitive marker for liver injury induced by RST in the early stage.
