摘 要:目的:探讨了连续性血液净化治疗对感染性休克猪TOLL样受体4表达水平的影响。方法:6只幼猪分别静脉注射Lps 100μg/kg,构建感染性休克模型。构建成功后4只感染性休克猪分别给予连续性血液净化治疗。采用流式细胞术检测治疗前和治疗后2、4、8h的感染性休克猪模型外周血单核细胞TLR4的表达水平,采用免疫组化检测治疗前和治疗后8h感染性休克猪模型肾组织中TLR4的表达水平。结果:与治疗前比较,治疗后幼猪外周血单核细胞TLR4的表达水平呈时间依赖性的下降,具有显著统计学意义(P<0.05)。与治疗前相比较,治疗后8h幼猪肾脏组织中TLR4的表达水平显著性下调,具有统计学意义(P<0.05)。结论:连续性血液净化治疗可改善免疫系统以及肾脏组织的功能,对重建机体免疫稳定具有重要作用。Objective:To explore expression of Toll like receptor 4 in septic shock pigs by continuous blood purification.Methods:Model of septic shock was established by injecting LPS(100μg/kg)into pigs.Septic shock pigs were treated by continuous blood purification.TLR4 expression of Peripheral blood mononuclear cell from treated septic shock pigs was determined by Flow cytometry.TLR4 expression of renal tissue from treated septic shock pigs was determined by Immunohistochemistry.Results: TLR4 expression level of peripheral blood mononuclear cell by time dependent reduced after treatment than before(P〈 0.05).TLR4 expression level of renal tissue at 8h after treatment significantly reduced than before(P〈0.05).Conclusion:Continuous blood purification therapy may improve the immune systemand kidney function.it plays an important role in rebuilding the immune stability.
