摘 要:目的观察吡格列酮对高脂饮食诱导的肥胖sD大鼠肝脏组织中脂联素表达的影响。方法选取sD大鼠40只,随机分为对照组(NC组)10只,给予普通饲料;高脂组(HF组)[包括:高脂对照组(HFN组)15只、高脂+吡格列酮组(HFP组)15只],给予高脂饲料,同时给予HFP组吡格列酮30mg·kg-1·d-1灌胃2周,Nc组和HFN组给予相同体积的0.9%氯化钠注射液灌胃,分别喂养12周后,测定大鼠体质量、空腹血糖、胰岛素、甘油三酯、总胆固醇水平,观察肝脏改变;用免疫组化法检测大鼠肝脏组织中脂联素的表达。结果12周后,HF组胰岛素敏感指数显著低于NC组[(0.021±0.010)与(0.015±0.007),P〈0.05],HFN组大鼠肝脏组织中脂联素,与NC组相比显著降低[(2929.73±157.45)与(3814.21±211.42),P〈0.05)],HFP组大鼠肝组织中脂联素的表达显著低于HFN组[(2929.73±157.45)与(3657.68±217.31),P〈0.05]。结论吡格列酮可升高肥胖大鼠模型肝组织中脂联素的表达,缓解胰岛素抵抗。Objective To observe the influence of pioglitazone on the expression of adiponectin in the liver of high fat diet-induced obese rats. Methods Forty male SD rats were randomly divided into control group which received regular diet( n = 10 ) ,high fat diet group( n = 15 ) and high fat diet goup administened pioglitazone( n = 15 ) on high fat diet. After twelve weeks, fasting blood glucose, fasting serum insulin, triglyceride, total cholesterol, and the histomorphological changes in liver were observed;insulin sensitivity test was performed;the fatty experimental group are treated with Pioglitazone( 30mg · kg- 1· d -1, oral garage) ,treatment was administered daily for 2 weeks. The control group and fatty control group all received an equal volume of vehicle (saline). The protein expression of adiponectin in the liver of rats was determined by immunohistochemistry. Results The insulin sensitive index was significantly decreased in the high-fat-diet rats compared with the control group [(0. 021 ± 0.010), (0. 015 ± 0. 007 ), P 〈 0.05 ] ; The protein level of adiponectin in the liver of fatty control group was significantly decreased compared with the control group [ (2929.73 ± 157.45 ), ( 3814.21 ± 211.42 ), P 〈 0.051, adiponectin in the liver of fatty experimental group was significantly increased compared with the fatty control group[ ( 2929.73 ± 157.45 ), ( 3657.68 ± 217.31 ), P 〈 0. 05 ]. Conclusion Pioglitazone could improve the expression of adiponectin, and improve the resistance of insulin.
