摘 要:从38个10bp寡聚核苷酸随机引物中筛选出20个扩增重复性好、条带清晰、特异性强、扩增带丰富的引物,对四川裂腹鱼、昆明裂腹鱼和灰裂腹鱼各12个个体基因组DNA进行随机扩增。每个引物在每个样本中扩增出6.3条带,扩增带大小在200-2200bp之间,在四川裂腹鱼、昆明裂腹鱼和灰裂腹鱼基因组DNA中分别检测到124、127和129个位点,多态位点分别为60个(占48.39%)、77个(占60.63%)和79个(占61.24%)。种间遗传距离显示:四川裂腹鱼和昆明裂腹鱼种间遗传距离指数为0.3979±0.0274,四川裂腹鱼和灰裂腹鱼种间遗传距离指数为0.3131±0.0286,昆明裂腹鱼和灰裂腹鱼种间遗传距离指数为0.4346±0.0328。结果表明,四川裂腹鱼和灰裂腹鱼之间的亲缘关系最近,昆明裂腹鱼和灰裂腹鱼亲缘关系最远。Gene complex of 12 individual commits of Schizothorax Kozlovi, Schizothorax Grahami and chizothorax Pellegrin were amplified by the 20 random primers, which were screened from 38 random primers with contained 10bp oligonueleotide nucleotide, and which were repeatable, strip clear and specific strong. After the amplification, 124 DNA fragments were produced in Schizothorax Kozlovi; 127 DNA fragments were produced in Sehizothorax Grahami; and 129 DNA fragments were produced in Schizothorax pellegrin. The polymorphic points were 60, 77 and 79 with the frequency of mean 48.39%, 60.63% and 61.24%. Between the Schizothorax Kozlovi and Schizothorax Grahami, the average genetic distance was 0.3979 ± 0.0274. Between the Schizothorax Kozlovi and Schizothorax pellegrin, the average genetic distance was 0. 3131 ±0. 0286. Between the Schizothorax Grahami and Sehizothorax pellegrin, the average genetic distance was 0. 4346 ± 0. 0328. The results show that the genetic relationship between the Schizothorax Kozlovi and Schizothorax pellegrin was the nearest, and the genetic relationship between the Schizothorax Grahami and Schizothorax pellegrin was the farthest.
