摘 要:目的研究木犀草素对UW液低温保存离体大鼠心脏的功能及对心肌细胞凋亡的影响。方法将40只成年SD大鼠随机分成2组(n=20):对照组(UW组)及实验组(添加30μmol/L木犀草素的UW液)。利用Langendorff离体心脏灌流法,观察离体心脏冷保存18h复灌60min后心率、左心室最大收缩压、左心室压力变化率、心肌细胞凋亡及凋亡相关基因Bax、Bcl-2的表达。结果与对照组比较,实验组复灌期心脏的收缩功能(LVPSP,+dp/dtmax)、舒张功能(-dp/dtmax)及心率高于对照组差异有统计学义意义(P<0.05);心肌细胞的凋亡及促进凋亡Bax基因的表达均明显低于对照组;而抑制凋亡Bcl-2基因的表达均明显高于对照组。结论木犀草素能显著改善UW液低温保存离体大鼠心脏的功能,对心肌细胞凋亡有明显抑制作用。Objective To study the effect of digitoflavone on isolated rat heart function and cell apoptosis after hypothermic preservation in University of Wisconsin solution(UW solution).Methods Forty male SD rats were randomly divided into 2 groups(n=20 in each group):control group and experiment group(added 30μmol/L digitoflavone in UW solution).Langendorff model of isolated rat heart was used.The cardiac contractile function and diastolic function were determined after 18 hours hypothermic preservation followed by 60-min reperfusion.The myocardial cell apoptosis and expression of Bax and Bcl-2 gene,which were related to apoptosis,were also observed.Results The results revealed that digitoflavone improved the recovery of cardiac contractile function(LVPSP,+dp/dtmax) and diastolic function(-dp/dtmax);the number of myocardial cell apoptosis and expression of Bax gene significantly reduced,and expression of Bcl-2 significantly increased in experiment group,compared with that in control group.Conclusion These results indicate that digitoflavone can significantly improve isolated rat heart function and reduce cell apoptosis after hypothermic preservation in UW solution.
