摘  要:目的探讨石见穿醇提取物体外对血管内皮细胞生物学行为的影响。方法采用人脐静脉内皮细胞(HUVECs)原代培养模型,通过MTT法观察石见穿醇提取物对HUVECs及人肝癌HepG2细胞增殖的影响;通过Transwell小室迁移、体外小管形成实验,观察石见穿醇提取物对HUVECs迁移、小管形成能力的影响。结果 12.5-200μg/ml的石见穿醇提取物作用于HUVECs48h,细胞增殖抑制率为17.73%-59.70%,明显高于其作用于人肝癌HepG2细胞的7.19%-39.66%(P<0.01);12.5-200μg/ml石见穿醇提取物作用HUVECs 12h后,迁移抑制率为1.83%-64.51%,且抑制作用与药物浓度呈正相关(r=0.940,P<0.05);12.5-200μg/ml石见穿醇提取物作用于HUVECs 24h后,形成管腔数目明显减少,管腔不完整甚至消失。结论石见穿醇提取物在体外能够明显抑制HUVECs增殖、迁移和小管形成,改变血管内皮细胞生物学行为。Objective To investigate the effects of the extract of salvia chinensis benth(SCB) on the biological behavior of vascular endiothelial cells. Methods Human umbilical vein endothelial cells (HUVECs) were cultured primarily in vitro. The effect of the alcohol extract of SCB on HUVECs and human hepatic carcinoma HepG2 cell was observed by MTT assay. Transwell cabin test and tube formation assay were used to observe the effect of the alcohol extract of SCB on the migration and tube formation ability of HUVECs. Results SCB 12. 5μg/ml to 200μg/ml inhibited the proliferation of HUVECs by 17. 73% to 59. 70%, which were higher than 7.19% to 39.66% when they acted onhuman hepatic carcinoma HepG2 cell (P〈0. 01). The inhibition rates of migration were 1.83% to 64.51% when HUVECs were treated with SCB 12. 5 μg/ml to 200μg/ml for 12 hours. The dose of SCB was positively correlated to the inhibition rate of migration(r= 0. 940, P〈0. 05). When HUVECs were treated with SCB (12.5 to 200μg/ml) for 24 hours, the tube number was reduced and lumen became unintegrity, even disappeared. Conclusion The alcohol extract of SCB could inhibit proliferation, migration and tube formation of HUVECs and change the biological behavior of vascular endothelial cells.
