摘  要:目的:研究长时间大强度运动对骨骼肌BMP6表达的影响,进一步阐明运动性低血色素的发生机制。方法:12只雄性Wistar大鼠体重(250±10)g,随机分为对照组和运动组,其中,对照组不运动,常规饲养;运动组进行4周、6 d/w、坡度为0、速度为30 m/min的递增负荷跑台训练。前2周每天训练1次,时间从1 min开始,后2周每天训练2次,每次递增2 min,最后1次训练时间达到71min。4周末取材。血细胞自动分析仪测定大鼠血红蛋白(Hb)等血常规;RT-PCR检测大鼠骨骼肌BMP6 mRNA和肝脏Hepcidin mRNA表达;原子分光光度计法测血清铁和骨髓铁含量。结果:(1)运动组Hb低于对照组(P<0.05);(2)运动组骨骼肌BMP6 mRNA表达高于对照组(P<0.05);两组肝Hepcidin mRNA表达无明显差异;(3)运动组骨髓铁贮存和血清铁均低于对照组(P<0.05,P<0.01)。结论:长时间大强度运动可引起大鼠骨骼肌BMP6表达增强,机体贮存铁和运载铁能力均降低,导致Hb合成减少,这可能是引发运动性低血色素的重要原因之一。Objective The purpose of this study was to investigate the effect of strenuous exercise on the expression of BMP6 mRNA in order to understand the mechanism of exercise-induced hypochrosis. Methods Twelve male Wistar rats were randomly divided into two groups:control group and exercise group. Rats in exercise group ran on a treadmill 2 min at 30 m/min,0% grade for 4 weeks,6 days per week. In the first 2 weeks,they ran once a day,beginning with 1 min, and increasing 2 min every day. From the third week, they ran twice a day, and increasing 2 min every day. The duration of last running reached 71min. Blood samples were collected by the end of experiment for determination of red blood cell count, Hb concentration, and white blood cells count. Serum iron,iron in marrow and the expression of BMP6 mRNA in skeletal muscle and hepatic hepcidin mRNA were determined after exercise. Results ( 1 )The level of Hb and RBC in the exercise group significantly decreased as compared with those in the control group(P 〈 0.05). (2)The expression of BMP6 mRNA in skeletal muscle significantly decreased in the exercise group as compared with the control group(P 〈 0.05),while there was no difference in the expression of hepatic hepcidin mRNA between the two groups. (3)Serum iron and iron in marrow of the exercise group significantly decreased as compared with the control group (P 〈 0.05 and P 〈 0.01 ,respectively). Conclusions Strenuous exercise reduces the ability of iron storage and transport,which may lead to reduce Hb synthesis and cause exercise-induced hypochrosis.
