携带肠道病毒71型中和线性表位重组流感病毒的构建

摘  要:目的获得能够携带肠道病毒71型(EV71)中和线性表位的重组流感病毒。方法通过PCR方法,在流感病毒HA基因中插入EV71中和线性表位序列,获得重组质粒;进行反向遗传学操作,构建重组流感病毒。结果获得了2个重组质粒pPolⅠ/Ⅱ/WSN-HA-SP55和pPolⅠ/Ⅱ/WSN-HA-SP70,并构建了能够分别携带EV71中和线性表位SP55和SP70的2株重组流感病毒,重组病毒3次传代血凝效价都保持在2^7和2^5HAU,具有较好的稳定性。结论通过反向遗传学操作,构建出携带Ev71中和线性表位的重组流感病毒,为研制预防手足口病重组流感病毒载体疫苗奠定基础。Objective To construct recombined influenza viruses with a neutralizing linear epitope of enterovirus 71 (EV71 ). Methods For getting recombined plasmids, the sequence of a neutralizing epitope of EV71was inserted into influenza virus hemagglutinin(HA) gene by PCR, and constructed recombined influenza viruses by reverse genetics. Results In this study, two recombined plasmids pPol I/lI/WSN-HA-SP55 and pPol I/II/WSN-HA-SP70 were constructed, and two recombined influenza viruses with a neutralizing linear epitopes SP55 and SP70 were obtained, respectively. In three passages, they had a good stability with 27 and 25 HAU. Conclusions By reverse genetics, two recombined influenza viruses have been constructed , and the two viruses will have a better developing foreground for using as candidate strains of live vaccine for preventing hand-foot-and-mouth disease.
