脂肪干细胞复合脂肪颗粒移植的实验研究

摘  要:目的探索脂肪来源干细胞(ADSCs)与脂肪颗粒复合移植对植入脂肪成活率的影响。方法将1ml通过吸脂术获得的纯化脂肪颗粒分别与1ml含有下列细胞浓度的PBS混合:①原代ADSCs1×10^6/ml(A组);②原代ADSCs2×10^6/ml(B组);③二代ADSCs2×10^6/ml(D组);④二代ADSCs2×10^7/ml(E组);对照组为单纯的脂肪颗粒和1mlPBS混合移植(C组)。随机注射移植于40只裸鼠背部皮下。移植12周后,取出移植物,进行如下测量:①大体观察;②重量和体积测定;③HE染色,观察细胞形态和小血管形成情况;④计算机图像分析测定血管密度;⑤统计学比较各实验组与对照组移植物体积、重量以及血管密度的差异。结果①湿重与体积:A、B、D、E组与C组之间差异有统计学意义(P〈0.01);A组与B、D、E组之间差异有统计学意义(P〈0.01);B、D、E组之间差异无统计学意义(P〉0.05);②血管密度:A、B、D、E组与C组间差异有统计学意义(P〈0.01);A、B、D、E各组间差异无统计学意义(Jp〉0.05)。结论原代和二代不同浓度的ADSCs与脂肪颗粒复合移植均可提高脂肪移植物成活率,相同浓度的原代与二代的ADSCs在促进脂肪颗粒成活上无显著差异,在一定范围内,随ADSCs浓度的增加,脂肪颗粒成活率也会相应提高;但是超过了一定范围,脂肪颗粒成活率不会随ADSCs浓度的增加而无限提高。Objective To investigate the effect of adipose-derived stem cells (ADSCs) of Passage 1 and 2 assisted lipotransfer on the influence of survival rate of fat granule. Methods 1 ml pure fat tissue acquired from liposuction was mixed with 1 ml PBS containing with the following cells: (①P1 ADSCs of 1 × 10^6/ml ( Group A) ; ②P1 ADSCs of 2 × 10^6/ml ( Group B) ; ③P2 ADSCs of 2 × 10^6/ml ( Group D) ; ④P2 ADSCs of 2 × 10^7/ml ( Group E) , and Group C was the control group which was the mixture of 1 ml pure fat tissue with 1 ml PBS. And then the mixture was injected randomly under the back skin of 40 nude mouse. The transplanted fat tissue was harvested at 12 weeks postoperative, the following measures were performed: ①Gross observation; ②Wet weight and volume; ③After HE staining, cell morphology and the angiogenesis were evaluated; ④The blood vessel density was analyzed through the computer image; ⑤Statistics analysis was conducted. Results ①Wet weight and volume of fat grafts: there was significant difference between Group A, B, D, E and Group C ( P〈0.01 ) ; and the difference between Group A and other three experimental groups was significant ( P〈 0.01 ), either. However, the differnce among Group B, D and E was not significant ( P〉 0.05). ②Blood vessel density : there was significant difference between Group A, B, D, E and Group C ( P〈 0.01 ) ; and the difference between the four experimental group was not significant ( P〉 0.05). Conclusion P1 and P2 ADSCs had similar effect on improving the survival rate of fat granule. And there was no obvious difference of the same concentration between P1 and P2 ADSCs on the effect of fat survival. To some extent, the survival rate could be enhanced with the increase of the cell concentration, but it could not increase indefinitely over a certain range.
