摘 要:人胎肝细胞裂解液经膜超滤,在分子量10~30kD组分中可检测出人肝细胞生长因子(hHGF)活性。hHGF为一热稳定的蛋白质或多肽类物质。它可特异地刺激肝来源细胞~3H-TdR掺入的增加,并且存在量效依赖关系,而对非肝来源细胞的DNA合成无刺激作用。hHGF的生物活性及理化性质与某些已知因子,如胰岛素、胰高血糖素、血小板来源的生长因子、表皮生长因子及增殖刺激因子等有所不同。After ultrafiltration of the lysate of human fetal liver cells, the existence of human hepatic stimulatory activity was disclosed in the fraction with molecular weight ranging from 10 to 30 kD. Human hepatocyte growth factor (hHGF) of human fetal liver origin possesses the specificity of stimulation of DNA synthesis in hepatocytes or hepatoma cells, but no ef- feet on other cells including human promyelocytic leukemic cells (HL-60 cell line), human erythroleukemic cells (K562 cell line), mouse fibroblast cells (3T3 cell line) or fresh fetal spleen and mesangial cells. Meanwhile, hHGF is nondialyzable, heat stable and resistant to the action of DNase, RNase or neuraminidase, but is deactivated by proteinase K and chymotrypsin. Based on our preliminary determinations, the peculiarities of hHGF isolated from human fetal liver are obviously different from those of other factors known to stimulate proliferation of hepatic cells, such as insulin, glucagon, platelet derived growth factor, epidermal growth factor and multiplication stimulating activity.
