摘  要:目的利用高场强磁共振弥散加权成像(DWI)对神经生长因子(NGF)防治缺血性脑损伤作用进行评价。方法健康成年家犬25只,随机分为治疗组20只,对照组5只,均行右侧大脑中动脉栓塞(MCAO)模型制备,栓塞后30 min,治疗组给予NGF,并于MCAO后6、24 h,7 d及3个月时分别取材。各组均在MCAO前后行DWI检查,切片行HE染色、免疫组织化学染色及电镜检查。结果治疗组犬各时间段神经功能受损症状相对较轻;对照组犬相应症状较重。治疗组24 h、7 d和3个月平均扩散系数(ADC)值下降幅度较小,回升时间较早;对照组相同时间点ADC值下降幅度较大,回升时间也较延后(P<0.01)。结论经NGF治疗的犬脑梗死模型在不同时间段的临床表现及DWI的信号强度较对照组轻;可根据DWI图像及ADC值进行NGF的疗效评价及预后评估。Objective To assess the role of high field MR diffusion-weighted image(DWI) in prevention and treatment of ischemic cerebral injury with NGF. Methods Twenty-five healthy adult dogs were randomly divided into treatment group (n = 20)and control group (n = 5). Thirty minutes after a right MCAO model was established, the animals in treatment group were treated with NGF. Tissue samples were taken 6 h, 24 h, 7 d and 3 months after the model was established. The animals underwent MR DWI before and after the model was established. Tissue samples were cut into sections which were observed under light and electron microscope with HE and immunohistochemical staining. Results The symptoms of nerve function impairment were relatively milder in treatment group than in control group at different time points. The decreased amplitute of average diffusion coefficient(ADC)was lower and increased faster in treatment group than in control group(P(0.01). The lower the amplitute of average ADC was in control group, the longer the time for the amplitude of ADC to increase was. Conclusion The clinical manifestations and the MR DWI signals of dog MCAO model are milder in treatment group than in control group at different time points. The effect of NGF on cerebral infarction and its prognosis can be assessed according to the MR DWI and ADC.
