摘  要:目的建立荡气颗粒中大黄结合蒽醌含量紫外分光光度测定方法。方法先采用三氯甲烷超声振荡提取除去游离蒽醌,残渣用盐酸水解得到结合蒽醌水解物;利用大黄蒽醌类化合物能与醋酸镁反应显色的原理,以显色剂为空白,测定样品溶液在517 nm波长处得吸收度。结果对照品大黄素含量在2.1~16.8 mg·L-1范围内,浓度与吸光度之间线性关系良好(r=0.9999);样品的平均回收率为98.6%,RSD%为1.2%(n=6)。结论紫外分光光度法适用于荡气颗粒中结合蒽醌的含量测定。Objective To establish a method for the determination of the combined anthraquinones of Dangqi Granule. Methods Tri- chloromethane was used to abstract free anthraquinone by ultrasonic vibration, hydrochloric acid was used to hydrolyze after drying. Ac- cording to the theory that colouring reaction will occur while anthraquinones in rhubarb react upon magnesium acetate, the absorption val- ue of the sample at 517 nm was measured to caculate content directly. Results The calibration curve of the absorbency was linear with the concentration of emodin in the range of 2.1 - 16.8 mg · L^- 1, ( r = 0.999 9). The average recovery of emodin was 98.6%, and RSD was 1.2% (n = 6). Conclusion The method can be applied to determine combined anthraquinones in Dangqi Granule.
