摘  要:目的:研究醛固酮合酶(CYP11B2)基因-344C/T多态性与慢性心力衰竭(chronic heart failure,CHF)及血浆醛固酮水平的关系。方法:采用多聚酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测96例CHF患者和160例健康对照的CYP11B2基因-344C/T多态性,应用ELISA方法检测血浆醛固酮水平,分析CYP11B2基因-344C/T多态性与CHF及醛固酮水平的关系。结果:CHF组和对照组基因型和等位基因频率比较差异均无统计学意义(χ2=2.50,P=0.287;χ2=2.31,P=0.129),CHF组TT、CT、CC三组基因型血浆醛固酮水平分别为(181.49±46.23)、(160.31±28.82)、(148.02±24.39)pg/ml,组间比较差异有统计学意义(TT与CC比较,t=1.99,P=0.029;TT+CT与CC比较,t=1.71,P=0.043)。结论:CYP11B2基因-344C/T多态性与慢性心力衰竭的发生不相关,CHF患者血浆醛固酮水平与CYP11B2基因-344C/T多态性相关。Objective: To study the relationship between CYP11B2 gene-344T/C polymorphism,chronic heart failure(CHF) and plasma aldosterone levels in CHF patients.Methods: CYP11B2 gene-344T/C polymorphism was analyzed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) in 256 subjects(96 patients with CHF and 160 controls),and the levels of plasma aldosterone in patients with CHF was assessed by ELISA.Results: No significant difference was found in genotype or allele frequency distribution between groups of control and CHF(χ^2=2.50,P=0.287;χ^2=2.31,P=0.129).The levels of plasma aldosterone in patients with CHF was(181.49±46.23)pg/ml in TT genotype class,(160.31±28.82)pg/ml in TC class and(148.02±24.39)pg/ml in CC class;TT vs.CC,t=1.99,P=0.029;TT + CT vs.CC,t=1.71,P=0.043.Conclusion: There was no association between the-344T/C polymorphism of CYP11B2 gene and CHF.The-344T/C polymorphism of CYP11B2 gene was associated with plasma aldosterone levels in patients with CHF.
