针药并用对单纯性肥胖大鼠血清瘦素及下丘脑NPY基因表达的影响

摘　　要：目的:观察针灸配合中药石连饮对肥胖大鼠血清瘦素、神经肽Y含量和下丘脑神经肽Y基因表达的影响,探讨针药并用治疗单纯性肥胖的机制。方法:Wistar雄性大鼠120只,随机分为普食组及高脂组,高脂组用高脂饲料喂养16周制作肥胖大鼠模型。将造模成功的60只大鼠按随机化原则分为:针药组、西药组、模型组。针药组针刺大鼠"后三里"和"中脘"穴,接通电针仪,并灌喂中药石连饮15 g/kg,每日1次,连续治疗14天。每10天测体重、体长并计算Lee'S指数。放射免疫法检测血浆NPY、血清瘦素含量;免疫组织化学法检测下丘脑瘦素受体表达。结果:与模型组比较,针药组血清瘦素含量降低(P<0.01),下丘脑瘦素受体表达增加(P<0.01);针药组血清NPY含量和下丘脑NPY基因表达比模型组降低(P<0.01)。结论:肥胖大鼠NPY含量及其基因表达的改善是针药并用减肥的作用机制之一。针药并用对肥胖机体下丘脑和血清中lePtin良性调节作用是改善瘦素抵抗以及调节异常内分泌代谢的作用机制之一。Objective：To observe the effect of acupuncture combined with traditional Chinese medicine on blood plasma leptin,neuropeptide Y contents and hypothalamic neuropeptide Y gene expression in simple obesity rats so as to explore the underlying mechanism of acupuncture combined with traditional Chinese medicine in treating simple obesity rats.Methods：A total of 120 male Wistar rats were randomly divided into normal control and high grease groups,and high grease group was established by feeding high fat forage for 16 weeks.The 60 rats which were made model blest were randomly divided into shilianyin plus electro-acupuncture groups,wester medicine group,model groups.Electro-acupuncture was applied to ＂Zusanli＂ and＂Zhongwan＂,and pour Shilianyin 15 g/kg,once daily,for 14 days.The body weight,body length and Lee’S index were measured once every 10 days.Blood plasma neuropeptide Y and Serum leptin contents were measured by radioimmunoassay.Leptin receptor expression was detected with immunohistochemistry.Results：In comparison with model group,the blood plasma neuropeptide Y contents and hypothalamic neuropeptide Y gene expression of shilianyin plus electro-acupuncture group decreased significantly（P〈0.01）.Conclusion：The improvement of neusopeptide Y contents and gene expression is the result of the mechanism of acupuncture with Chinese medicine.The optimum accommodation role of acupuncture combined with traditional Chinese medicine to Hypothalamus and Serum leptin of obesity body is one of the mechanism of action to improve leptin-counteract and accommodate abnormal internal hormone metabolism.
