摘 要:目的 探讨丹参对急性肺损伤(ALI)的预防、治疗作用.方法 32 只SD 大鼠随机分为模型组、对照组、丹参预处理及丹参干预组,每组8 只.鼠尾静脉分别注射生理盐水、脂多糖及丹参.造模6h后,观察肺组织形态学改变,测定左肺/ 体重比值,免疫荧光法测定肺组织中核转录因子-κB(NF-κB)、炎症因子(IL-6、TNF-α)、细胞间黏附分子-1(ICAM-1).结果 肺组织形态学显示模型组肺组织明显充血、水肿,并有炎性细胞浸润,而在丹参预处理组及丹参干预组上述表现明显减轻;肺/ 体重比值模型组[(5.12±0.25)mg/g]较对照组[(4.65±0.45)mg/g]明显升高,但丹参预处理组[(4.80±0.15)mg/g]及丹参干预组[(4.78±0.29)mg/g]较模型组明显降低(P<0.01 或0.05);炎性因子(IL-6、TNF-α)、NF-κB 及ICAM-1 模型组较对照组分泌增多,但丹参预处理组及丹参干预组较模型组降低,且上述因子的分泌主要分布在血管内皮细胞附近.结论 丹参对ALI 起着预防和治疗作用,这种保护作用可能与抑制ICAM-1活性及NF-κB 信号转导途径有关.Objective To investigate the protective effects of Salvia injection against acute lung injury (ALl) in rats. Methods Thirty two SD rats were randomly divided into 4 groups: model group, control group, Salvia pretreatment and Salvia treatment group (n=8 in each group). The lung injury was induced by tail vein injection of lipopolysaccharide (LPS), in control group the normal saline was injected instead of LPS; the salvia injection was give before (pretreatment group) or after (treatment group) model was made. The lung tissue morphology changes were observed, the lung/body weight ratio was calculated, and the nuclear transcription factor-kappa B(NF-kappa B), inflammatory factors(IL-6, TNF-alpha), intercellular adhesion molecule-1 (I- CAM-l) of lung tissue were detected by immunofluorescence method. Results The lung tissue showed hyperemia, edema and inflammatory cell invasion in the model group, while the changes in Salvia pretreatment and Salvia treatment group were signifi- cantly attenuated. Lung/body weight ratio of model group [(5.12 + 0.25)mg/g] was higher than that of control group [(4.65 ~ 0.45)mg/g], salvia pretreatment group[(4.80 ~ 0.15) mg / g] and salvia treatment group [ (4.78 ~ 0.29) mg / g ] (P〈0.01 or 0.05). The immunofluoresence results showed higher levels of inflammatory factors (IL-6, TNF-alpha), NF-kappa B and ICAM-1 in model group than those in control group, salvia pretreatment and treatment groups; these factors were mainly distributed in the vicinity of vascular endothelial cells. Conclusion Salvia injection has a preventive and therapeutic effect on acute lung injury in rats, which may be related to the inhibition of ICAM-1 activity and NF- K B signal transduction pathway.
