摘  要:本实验研究从地衣芽孢杆菌发酵液中分离提取杆菌肽纯品。用生长12h的地衣芽孢杆菌作为种子菌,以10%接种量接种到发酵培养基中,37℃,220rpm/min发酵54.5h,8000rpm/min离心20min得上清液。萃取除去发酵液中的色素,用丁醇与发酵液以1:2体积比重复萃取2次,再将萃取的丁醇溶液与水以1:1体积比混合进行反萃取,重复2次得到萃取液,其萃取率为90.04%;树脂进行浓缩,筛选得到XH-8树脂效果良好,用55%酒精洗脱,洗脱液将溶液浓缩7.5倍。The study was related to the recovery of bacitracin from fermentation beer.Bacillus licheniformis which was growing 12h was used as seed bacteria,with 10% inoculum inoculated into fermentation medium,37℃,220rpm/min fermentation 54.5h.Then the fermentation beer was centrifuged at 8000rpm/min of 20min,obtaining the centrifugal liquid.Firstly,the pigment was removed by extraction.The fermentation beer and butanol was used for extraction with the 2:1 volume ratio.The fermentation beer layer was re-extracted once again.Secondly,the water and butanol was mixed with the 1:1 volume ratio.The butanol layer was re-extracted once again.And 90.04% of the bacitracin was obtained.Then the bacitracin was concentrated by resin,which adsorbed by the XH-8 resin and watered by 55% ethanol.Then the volume of extraction beer was 7.5 times of the watered beer we obtained.
