柯萨奇B_4病毒体内外感染所致小鼠胰岛B细胞的损伤

摘　　要：目的 :研究我国流行的柯萨奇B4 病毒亲胰腺株 (pancreatotropicstrainofCoxsackievirusB4 ,P -CB4 V)感染与胰岛素依赖型糖尿病 (insulin -dependentdiabetesmellitus,IDDM )之间的关系。 方法 :P -CB4 V经体内外感染Swiss小鼠后 ,测定血糖及糖刺激的胰岛素释放量。结果 :80 %的小鼠在感染后 72h出现低血糖 ,血糖在(90 87± 8 2 6 )mg/L范围之内 ,6～ 8wk全部发展为高血糖。糖刺激的胰岛素释放量在感染后 72h增加 ,但至 3～ 8wk明显低于未感染组。同时 ,用P -CB4 V对体外培养的小鼠胰岛及胰岛细胞进行攻击 ,经低糖及高糖刺激的胰岛素释放量均明显低于对照组。结论 :P -CB4 V经体内或体外感染 ,均可损伤小鼠B细胞。Objective:To study the relationship between viral etiology of IDDM and pancreatotropic strain of Coxsackie B 4 virus (P-CB 4V), which is epidemic in China.Methods:(1)Swiss mice infected with P-CB 4V in vivo and in vitro. (2) Blood glucose test. (3)Tests of insulin secretion of islets of infected mice after stimulation by glucose. Results:Hypoglycemia appeared in 80% of the mice at 72h pi. with blood sugar level at (90.87±8.26)mg/L, and hyperglycemia in total mice from 6 to 8wk pi.Insulin secretion level was increased after stimulation of sugar at 72h p i, however, it was decreased from 3 to 8wk pi, as compared with controlled group. Meanwhile, the cultured islets and B cells attacked with the P-CB 4V in vitro. The insulin secretion level was lower remarkably compared with controlled group. Conclusion:The damage of mouse islets cells can be made with CB 4V infection both in vivo and in vitro.
