摘 要:通过对红王子锦带花粉胚发育的细胞学观测与统计分析,对适宜红王子锦带花粉启动分裂的因子、适宜的花粉发育时期、适宜的接种时期以及适宜的培养基组合等条件进行了筛选。结果表明:适宜花粉启动分裂的因子为6-BA与MS基本培养基,6-BA的浓度越高,MS的浓度越低,出胚率越高;适宜花粉启动分裂的发育时期为四分体时期,出胚率为6.73%;适宜诱导的接种时期为6月;适宜的培养基组合为:1/3MS+6-BA2.0mg.L-1+NAA1.0mg.L-1+1%SUC。Through cytologic observation and statistical analysis on the pollen embryos of Weigela florida cv. 'Red Prince' , the factors of suitable pollen starting division,suitable pollen development stage, suitable inocula- ting stage,suitable medium combination and other conditions were screened out. The results of showed that the main factors of suitable pollen starting division were 6-BA and MS basic medium,the higher level of 6-BA and the lower level of MS basic medium, the higher rate of embryos;The suitable development stage of pollen start- ing division was tetrad stage,the rate of embryos was 6.73% ;The suitable inoculating stage induction was in June;The suitable inducing medium was 1/3MS+6-BA2.0mg·L-1+NAAl.0mg·L-1+1%SUC.
